目的：验证LncRNA UCA1与miRNA‐99b的相关性。方法：采用荧光定量PCR法检测UCA1和miRNA‐99b在过表达和敲低表达 UCA1的膀胱癌细胞、膀胱癌组织中的表达。结果：在敲低 UCA1表达的膀胱癌细胞5637中，UCA1表达下调60％，而miRNA‐99b的表达上调250％；在过表达UCA1的膀胱癌细胞 UM UC2中，UCA1表达上调410％，而miRNA‐99b的表达下调60％；UCA1在膀胱癌中的表达水平明显高于癌旁组织（P＜0．05），而miRNA‐99b在膀胱癌中的表达水平明显低于癌旁组织（P＜0．05）；UCA1与miRNA‐99b在膀胱癌组织中的表达呈负相关（R＝‐0．502，P＜0．05）。结论：UCA1负向调节miRNA‐99b的表达，但是它对miRNA‐99b的调节在膀胱癌发生发展中的作用机制还需深入研究。
목적：험증LncRNA UCA1여miRNA‐99b적상관성。방법：채용형광정량PCR법검측UCA1화miRNA‐99b재과표체화고저표체 UCA1적방광암세포、방광암조직중적표체。결과：재고저 UCA1표체적방광암세포5637중，UCA1표체하조60％，이miRNA‐99b적표체상조250％；재과표체UCA1적방광암세포 UM UC2중，UCA1표체상조410％，이miRNA‐99b적표체하조60％；UCA1재방광암중적표체수평명현고우암방조직（P＜0．05），이miRNA‐99b재방광암중적표체수평명현저우암방조직（P＜0．05）；UCA1여miRNA‐99b재방광암조직중적표체정부상관（R＝‐0．502，P＜0．05）。결론：UCA1부향조절miRNA‐99b적표체，단시타대miRNA‐99b적조절재방광암발생발전중적작용궤제환수심입연구。
Objective :To confirm the correlation of LncRNA UCA1 and miRNA‐99b .Methods :UCA1 and miRNA‐99b were assessed in bladder carcinoma celles with over expression and knockdown of UCA 1 ,and bladder carcinoma tissues .Results :UCA1 was down‐regulation by 60% ,while miRNA‐99b was up‐regulation by 250% in bladder carcinoma cell 5637 with knockdown of UCA1 .UCA1 was up‐regulation by 410% ,while miRNA‐99b was down‐regulation by 60% in bladder carcinoma cell UMUC2 with over expression of UCA1 .UCA1 was significantly increased in bladder carcinoma tissues compared with adjacent carcinoma tissues (P<0 .05) ,but miRNA‐99b was completely opposite(P<0 .05);The correlation of UCA1 and miRNA‐99b in bladder carcinoma tissues was analy‐sised(R=‐0 .502 ,P<0 .05) .Conclusion:UCA1 is negative regulating the expression of miRNA‐99b ,but it's to be re‐searched in further of the correlation of UCA1 and miRNA‐99b in the mechanism of bladder tumorous development .
