为探究欧美杂交杨转录调控因子 Pnd-WRKY3在落叶松-杨栅锈菌( Melam pos ra larici-populina) E4生理小种侵染欧美杨(Populsu nigra×P.deltoides)后不同时间点的具体功能,根据转录组测序结果设计特异性引物进行Pnd-WRKY3基因CDS区克隆,应用生物信息学预测Pnd-WRKY3调控的靶基因,采用荧光定量Q-PCR研究Pnd-WRKY3基因抗锈菌表达特性。结果表明:Pnd-WRKY3基因蛋白编码区全长1779 bp,预测具有W盒的靶基因49条,其中杨树动力蛋白表达量与Pnd-WRKY3基因表达量正相关。 Q-PCR 结果表明,锈菌菌丝生长期及生物量大量增长时期,Pnd-WRKY3基因表达量上调。在锈菌完成一个生长周期形成夏孢子时,Pnd-WRKY3基因表达量开始下降。

In order to reveal the resistance molecular mechanism of the hybrid poplar Robusta ( Popul us nigra×P.deltoides) to the E4 virulent filial generation of Melampsora laric-populina, we cloned the Pnd-WRKY3 transcription factor gene , and studied the gene function of Pnd-WRKY3 during the rust infection .The complete CDS of Pnd-WRKY3 was 1 779 bp.Then, we predicted 49 target genes of Pnd-WRKY3 with the W box, including one dynamin protein gene , in which expression quantity had positive correlation with that of Pnd-WRKY3.By Q-PCR, the expression of Pnd-WRKY3 was promoted during rust mycelial growth period and the reproductive period .The expression was suppressed when spores were formed , then the life cycle of rust was finished at the same stage .
