He Shen pill%Adriamycin - induced nephropathy%Podocytes
目的:利用阿霉素肾病模型研究禾肾丸对大鼠肾脏形态学的影响。方法实验动物分为空白组、模型组和禾肾丸组。采用一次性尾静脉注射阿霉素6 mg/ kg,复制阿霉素肾病大鼠模型,造模成功后每日用禾肾丸灌胃,空白组及模型组以蒸馏水灌胃。(1)在造模第2周及8周处死大鼠,观察各组大鼠肾脏情况,收集24 h 尿液检测尿蛋白;(2)造模第2周及第8周取各组大鼠肾组织进行光镜观察;(3)取肾脏组织电镜下观察肾小球足细胞足突、基底膜变化。结果(1)肉眼观:禾肾丸组大鼠肾脏较模型组质地偏软,颜色鲜红,体积偏大,各组大鼠尿蛋白检测提示禾肾丸组较模型组减少;(2)通过采用 HE、Masson 染色观察各组大鼠肾脏组织形态学的改变,系膜细胞增生情况、肾小管-间质病变情况,模型组在第4周开始出现系膜细胞增生,第6周开始出现肾小球局灶节段的硬化,而禾肾丸组硬化程度较模型组轻;(3)电镜显示禾肾丸组肾脏足细胞足突融合较模型组减少,裂孔膜间隙小。结论禾肾丸对于阿霉素肾病模型具有治疗作用,保护肾小球足细胞,减轻阿霉素肾病大鼠肾组织纤维化程度。

목적:이용아매소신병모형연구화신환대대서신장형태학적영향。방법실험동물분위공백조、모형조화화신환조。채용일차성미정맥주사아매소6 mg/ kg,복제아매소신병대서모형,조모성공후매일용화신환관위,공백조급모형조이증류수관위。(1)재조모제2주급8주처사대서,관찰각조대서신장정황,수집24 h 뇨액검측뇨단백;(2)조모제2주급제8주취각조대서신조직진행광경관찰;(3)취신장조직전경하관찰신소구족세포족돌、기저막변화。결과(1)육안관:화신환조대서신장교모형조질지편연,안색선홍,체적편대,각조대서뇨단백검측제시화신환조교모형조감소;(2)통과채용 HE、Masson 염색관찰각조대서신장조직형태학적개변,계막세포증생정황、신소관-간질병변정황,모형조재제4주개시출현계막세포증생,제6주개시출현신소구국조절단적경화,이화신환조경화정도교모형조경;(3)전경현시화신환조신장족세포족돌융합교모형조감소,렬공막간극소。결론화신환대우아매소신병모형구유치료작용,보호신소구족세포,감경아매소신병대서신조직섬유화정도。
Objective To study the effect of Heshen pills on glomerular podocytes in adriamycin -induced nephropathy model rats. Methods SD rats were divided into three groups,including the normal group,the model group,and the Heshen pill group.(1)On the second,4th,6th,and 8th week of modeling,the urine sample was collected to examine the dynamic changes of 24 hours urinary protein quantity.(2)On the second and 8th week after modeling,the changes of renal tissues were observed under light microscope.(3) Renal tissue were observed under electron microscope for changes of glomerular podocyte foot process and basement membrane. Results (1)Compared with model group,according to the naked eye view,Heshen group rats showed texture soft,bright red,big volume kidney,and urine protein test indicated decrease;(2) HE and Masson staining were applied to observe each group in the rat kidney tissue morphology change,me-sangial cell proliferation situation,renal tubular interstitial lesions. The model group began to appear mesang-ial cell hyperplasia on the 4th week,and visualized focal segmental glomerular sclerosis showed on the 6th week. The degree of hardening of rats in Heshen pill group was lighter;(3)Electron microscopy showed that Heshen group had less kidney podocyte foot process fusion compared with model group with smaller clear-ance. Conclusion Heshen pills has curative effects on adriamycin nephrosis model,and it can protect glo-merular podocyte,reduce adriamycin nephrosis rats renal tissue fibrosis degree.
