目的:探讨决明子乙醇提取物(Semen Cassiae Extract,SCE)对高脂高糖饮食所致的非酒精性脂肪肝大鼠调脂保肝作用。方法 SD大鼠72只,雌雄各半,随机分成正常对照组(12只)和模型组(60只)。正常对照组给予普通饲料,饮用蒸馏水,模型组大鼠给予高脂饲料,饮用10%果糖水。饲养至第6周末,将模型组大鼠随机分为模型对照组(DW10mL· kg-1)、二甲双胍组(0.2g· kg-1)、决明子乙醇提取物(SCE)高(2g· kg-1)、中(1g· kg -1)、低(0.5g· kg -1)剂量组。连续灌胃给药4周后,测定大鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、谷胱甘肽-S转移酶( GST)活性,血清低密度脂蛋白胆固醇( LDL-C)、高密度脂蛋白胆固醇( HLD-C)、总胆固醇( TC)、甘油三酯( TG)含量。测定大鼠肝脏组织TC、TG含量。测定大鼠的肝脏指数,并作HE染色观察病理组织学变化。结果与正常对照组相比,模型组大鼠血清TC、TG和 LDL-C的含量明显升高(P<0.01),HLD的含量明显降低(P<0.01);血清内AST、ALT和GST的含量明显升高(P<0.01),肝脏损伤指数增加,肝细胞内广泛性脂肪浸润。 SCE和二甲双胍处理4周后,均有不同程度的下调模型大鼠血清TC、TG和LDL-C的含量,升高HLD的含量( P<0.05,P<0.01);血清内AST、ALT和GST的含量有不同程度的下降( P<0.01,P<0.05),降低肝脏损伤指数,肝组织脂肪浸润明显减少,肝窦清晰,肝索排列整齐。结论决明子提取物对脂肪肝大鼠具有调脂保肝作用。

목적:탐토결명자을순제취물(Semen Cassiae Extract,SCE)대고지고당음식소치적비주정성지방간대서조지보간작용。방법 SD대서72지,자웅각반,수궤분성정상대조조(12지)화모형조(60지)。정상대조조급여보통사료,음용증류수,모형조대서급여고지사료,음용10%과당수。사양지제6주말,장모형조대서수궤분위모형대조조(DW10mL· kg-1)、이갑쌍고조(0.2g· kg-1)、결명자을순제취물(SCE)고(2g· kg-1)、중(1g· kg -1)、저(0.5g· kg -1)제량조。련속관위급약4주후,측정대서혈청중곡병전안매(ALT)、곡초전안매(AST)、곡광감태-S전이매( GST)활성,혈청저밀도지단백담고순( LDL-C)、고밀도지단백담고순( HLD-C)、총담고순( TC)、감유삼지( TG)함량。측정대서간장조직TC、TG함량。측정대서적간장지수,병작HE염색관찰병리조직학변화。결과여정상대조조상비,모형조대서혈청TC、TG화 LDL-C적함량명현승고(P<0.01),HLD적함량명현강저(P<0.01);혈청내AST、ALT화GST적함량명현승고(P<0.01),간장손상지수증가,간세포내엄범성지방침윤。 SCE화이갑쌍고처리4주후,균유불동정도적하조모형대서혈청TC、TG화LDL-C적함량,승고HLD적함량( P<0.05,P<0.01);혈청내AST、ALT화GST적함량유불동정도적하강( P<0.01,P<0.05),강저간장손상지수,간조직지방침윤명현감소,간두청석,간색배렬정제。결론결명자제취물대지방간대서구유조지보간작용。
ABSTRACT:OBJECTIVE To investigate the protective and lipid-decreasing effects of Semen Cassiae Extract (SCE) on fatty liver in rats.METHODS 72 SD rats were randomly divided into normal group (12) and model group (60).The normal rats were raised by standard animal diet ,while the model rats were fed with high-fat diet and 10%fructose for 6 weeks.Model rats were randomly divided into 5 groups,including fatty liver group,metformin group (0.2g· kg -1),SCE high-dose group (2g· kg -1),middle-dose group(1g· kg -1) and low-dose group (0.5g · kg -1 ).All rats were treated for 4 weeks with drugs.The contents of ALT,AST,GST,TG,TC,HDL-C,LDL-C in the serum and TG ,TC in the liver tissue were respectively measured.Liver index was measured.The left leaf of liver was observed by histopathological staining.RESULTS Compared with normal rats ,the contents of ALT ,AST,GST, TG,TC in the serum and TG ,TC in the liver tissue were increased ( P<0.01 ) ,but the contents of HDL-C were de-creased (P<0.01) in model rats.After SCE treatment for 4 weeks,compared with model rats,the contents of ALT, AST,GST,TG,TC in the serum and TG,TC in the liver tissue were decreased respectively ,but the content of HDL-C was increased.According to the histopathological staining ,SCE remarkably lighten inflammatory cell infiltration and fibrosis increasing.CONCLUSION SCE possess protective and lipid-decreasing effect on fatty liver in rats.
