Shipi gushen huayu recipe%Adriamycin%Renal fibrosis
目的:探讨实脾固肾化瘀方对阿霉素(ADR)肾纤维化大鼠肾功能的保护作用及其可能的机制。方法:将雄性 SD 大鼠40只随机分为4组(正常组、模型组、治疗组、对照组),除正常组外,其余大鼠均采用尾静脉注射阿霉素(4 mg/ kg)法制成 ADR 肾纤维化大鼠模型。造模后2周,治疗组予实脾固肾化瘀方浸膏(43 g/ kg)灌胃;对照组予福辛普利钠溶液(2 mg/ kg)灌胃;正常组和模型组予生理盐水灌胃,1/ d。观察30 d 后留取大鼠24 h 尿蛋白定量,取血处死并取肾脏组织,观察各组Ⅰ型胶原(Col Ⅰ)、层黏蛋白(LN)、转化生长因子-β1(TGF -β1)及α-平滑肌肌动蛋白(α- SMA)表达变化。结果:治疗组大鼠肾组织 Col Ⅰ、LN、TGF -β1及α- SMA 表达较模型组明显下降(P ﹤0.01),与对照组比较差异无统计学意义(P ﹥0.05)。结论:实脾固肾化瘀方能显著改善 ADR 肾病大鼠肾纤维化,保护肾功能,减轻肾功能损害,与福辛普利钠比较,前者更具有优势。

목적:탐토실비고신화어방대아매소(ADR)신섬유화대서신공능적보호작용급기가능적궤제。방법:장웅성 SD 대서40지수궤분위4조(정상조、모형조、치료조、대조조),제정상조외,기여대서균채용미정맥주사아매소(4 mg/ kg)법제성 ADR 신섬유화대서모형。조모후2주,치료조여실비고신화어방침고(43 g/ kg)관위;대조조여복신보리납용액(2 mg/ kg)관위;정상조화모형조여생리염수관위,1/ d。관찰30 d 후류취대서24 h 뇨단백정량,취혈처사병취신장조직,관찰각조Ⅰ형효원(Col Ⅰ)、층점단백(LN)、전화생장인자-β1(TGF -β1)급α-평활기기동단백(α- SMA)표체변화。결과:치료조대서신조직 Col Ⅰ、LN、TGF -β1급α- SMA 표체교모형조명현하강(P ﹤0.01),여대조조비교차이무통계학의의(P ﹥0.05)。결론:실비고신화어방능현저개선 ADR 신병대서신섬유화,보호신공능,감경신공능손해,여복신보리납비교,전자경구유우세。
Objective:To observe renal function protective effects of Shipi Gushen Huayu Recipe on adriamycin(ADR)in-duced renal fibrosis in rats and its probable mechanism. Methods:40 male SD rats were randomly divided into four groups(normal group,model group,treatment group and control group),in addition to normal group,the rest rats were created to renal fibrosis mod-el with adriamycin(4 mg/ kg)by tail intravenous injection. Treatment group were administered by Shipi Gushen Huayu Recipe extract (43 g/ kg)after creation of model for 2 w;Control group were administered by fosinopril solution(2 mg/ kg);Both normal group and model group were administered by saline one time a day. After 30 days,all rats were examined 24 hours urine protein quantity,blood were collected before sacrificed,and kidney tissue were remained to observe the expression of Col Ⅰ(Collagen Ⅰ),LN(Laminin), TGF - β1(Transfer growth factor - β1 )and α - SMA(α - Smooth muscle albumin)in treatment group. Results:Expression of ColⅠ,LN,TGF - β1 and α - SMA in treatment group were significantly decreased than that in model group(P ﹤ 0. 01). There was no statistically significant difference compared with control group(P ﹥ 0. 05). Glomerular mesangial cells and basement membrane hy-perplasia in treatment group were improved significantly than that in model group under light microscope. Conclusion:Shipi Gushen Huayu Recipe can significantly improve ADR induced renal fibrosis in rats,alleviate renal function impairment,compared to fosinopril sodium the former has more advantages.
