目的探讨人脐血单个核细胞(HCMNCs)对半乳糖卵巢早衰小鼠动情周期及雌二醇(E2)、卵泡刺激素(FSH)、黄体生成激素(LH)的影响,评价卵巢功能的恢复情况。<br> 方法将120只雌性 SPF级昆明小鼠随机分为 A(半乳糖卵巢早衰+ HCMNCs组)、B(半乳糖卵巢早衰组)、C(空白对照组)三组,每组40只。采用给昆明小鼠饲喂35%半乳糖建立半乳糖卵巢早衰小鼠动物模型,聚蔗糖密度梯度离心法制备 HCMNCs悬液。造模成功后,A组小鼠双侧卵巢内分别注入10μl HCMNCs悬液;B组小鼠双侧卵巢内注入等量 L-DMEM培养液;C组未行任何处理。移植后50 d,ELISA法测定昆明小鼠血清雌二醇(E2)、卵泡刺激素(FSH)及黄体生成激素(LH)变化情况。<br> 结果 HCMNCs移植前,A、B组与 C组比较,动情周期紊乱率升高、血清 E2降低、FSH及 LH升高,差异均有统计学意义(P <0.05)。HCMNCs移植后,A组动情周期紊乱率下降、血清 E2升高,FSH及 LH降低,与移植前比较差异均有统计学意义(P <0.05);A组与 C组移植后比较,动情周期紊乱率、E2、FSH及 LH无明显差异(P >0.05);A、C组与 B组移植后比较,动情周期紊乱率、E2、FSH及 LH有统计学差异(P <0.05)。<br> 结论 HCMNCs能促进半乳糖卵巢早衰小鼠卵巢功能的恢复。

목적탐토인제혈단개핵세포(HCMNCs)대반유당란소조쇠소서동정주기급자이순(E2)、란포자격소(FSH)、황체생성격소(LH)적영향,평개란소공능적회복정황。<br> 방법장120지자성 SPF급곤명소서수궤분위 A(반유당란소조쇠+ HCMNCs조)、B(반유당란소조쇠조)、C(공백대조조)삼조,매조40지。채용급곤명소서사위35%반유당건립반유당란소조쇠소서동물모형,취자당밀도제도리심법제비 HCMNCs현액。조모성공후,A조소서쌍측란소내분별주입10μl HCMNCs현액;B조소서쌍측란소내주입등량 L-DMEM배양액;C조미행임하처리。이식후50 d,ELISA법측정곤명소서혈청자이순(E2)、란포자격소(FSH)급황체생성격소(LH)변화정황。<br> 결과 HCMNCs이식전,A、B조여 C조비교,동정주기문란솔승고、혈청 E2강저、FSH급 LH승고,차이균유통계학의의(P <0.05)。HCMNCs이식후,A조동정주기문란솔하강、혈청 E2승고,FSH급 LH강저,여이식전비교차이균유통계학의의(P <0.05);A조여 C조이식후비교,동정주기문란솔、E2、FSH급 LH무명현차이(P >0.05);A、C조여 B조이식후비교,동정주기문란솔、E2、FSH급 LH유통계학차이(P <0.05)。<br> 결론 HCMNCs능촉진반유당란소조쇠소서란소공능적회복。
Objective To explore the effect of the human umbilical cord blood mononuclear cells (HCMNCs) on estrous cycle of premature ovarian failure induced by galactose in mice, estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH) and evaluate the recovery of ovarian function. <br> Methods 120 female SPF Kunming mice were randomly divided into three groups: A (premature ovarian failure induced by galactose + HCMNCs, n = 40), B (premature ovarian failure induced by galactose group, n = 40), C (control group, n = 40). The model of premature ovarian failure in mice was established by feeding Kunming mice with 35% galactose HCMNCs suspension was prepared by the method of density gradient centrifugation. The mouse ovaries in group A were injected with 10μl HCMNCs suspension per mouse, ones in group B were done with equivalent medium, and group C without any treatment. The changes of serum E2, FSH and LH in Kunming mice were determined by ELISA method for 50 d after transplantation. <br> Results Before the transplantation of HCMNCs, the disorder rate of the estrous cycle in group C was significantly increased, and the serum E2 was clearly decreased, but FSH and LH were increased as compared to group A or group B. There existed statistically significant differences (P < 0.05). After HCMNCs were transplanted, the level of LH and FSH in the A group, was decreased and serum E2 was increased. There were statistically significant differences for group A as compared to ones before the transplantation of HCMNCs (P < 0.05). The disorder rate of estrous cycle, LH, FSH and E2 were not significantly changed for group A or group C with/without transplantation (P > 0.05). However, the disorder rate of estrous cycle, the level of E2, FSH and LH in group B was significantly different from ones in group A or group B after transplantation (P < 0.05). <br> Conclusion HCMNCs can improve the recovery of ovarian function of premature ovarian failure induced by galactose in mice.
