目的:探讨结直肠癌组织中HIF-1α与拓扑异构酶-Ⅱ(Topo-Ⅱ)的表达变化,及其对奥沙利铂化疗患者预后的影响。方法采用免疫组化SP法检测HIF-1α、Topo-Ⅱ在结直肠癌组织中的表达情况,了解其对化疗疗效的影响。结果 HIF-1α在大肠癌组织的表达阳性率为57.6%,明显高于癌旁和正常组织(<0.01);大肠癌组织中HIF-1α和Topo-Ⅱ的表达呈正相关,Spearman相关系数为0.247,<0.05。结论结直肠癌组织中HIF-1α的表达与肿瘤侵润深度、原发灶是否伴有淋巴结转移有关,HIF-1α可以促进肿瘤的侵袭及转移能力,HIF-1α、Topo-Ⅱ可能在肿瘤多药耐药机制、判断大肠癌转移和耐药性方面具有一定的协同作用。

Objective To detect the expression of HIF-1α, Topo-II, and to investigate the cor elation between the HIF-1α, Topo-II expression with the ef icacy of adjuvant oxaliplatin based chemotherapy in colorectal cancer patients.Methods The expression of HIF-1α and Topo-II were detected by immunohistochemical features in surgical excision specimens. Results HIF-1α was expressed in colorectal cancer tissues with the positive rate of 57.6% ,the positive expression rate of HIF-1αin colorectal carcinoma is significantly higher than the positive expression rate in the paraneoplastic and normal tissues, <0.01;In colorectal cancer, the expression of HIF-1αwas positively cor elated with that of Tope-II, and it was statistical y significant(r=0.247, <0.05). Conclusion HIF-1αand Tope-IImay have certain common function in tumor MDR mechanism, the combined use may has a synergistic inhibitory ef ect on colorectal cancer metastasis and resistance, helps early diagnosis of colorectal cancer and potential judgment of lymph node metastasis.
