目的 评价线粒体钙单向转运体在大鼠肺缺血再灌注损伤中的作用.方法 健康成年雄性Wistar大鼠30只,体重250~ 300 g,采用随机数字表法分为3组(n=10):假手术组(S组)、肺缺血再灌注组(I/R组)和线粒体钙单向转运体抑制剂钌红组(RR组).I/R组和RR组采用夹闭左肺门45 min,恢复灌注2h的方法制备肺缺血再灌注损伤模型;RR组于夹闭左肺门前1h时尾静脉注射钌红2.5 mg/kg.再灌注2h时,采集动脉血样0.5 ml,行血气分析,计算氧合指数;然后处死大鼠,取左肺上叶组织,计算肺组织湿重/干重(W/D)比值.结果 与S组比较,I/R组和RR组氧合指数降低,肺组织W/D比值升高(P<0 05);与I/R组比较,RR组氧合指数升高,肺组织W/D比值降低(P<0.05).结论 大鼠肺缺血再灌注损伤的机制与促进线粒体钙单向转运体的开放有关.

Objective To evaluate the role of mitochondrial calcium uniporter in lung ischemiareperfusion (I/R) injury in rats.Methods Thirty healthy male Wistar rats, weighing 250-300 g, were randomly divided into 3 groups (n=10 each) using a random number table: sham operation group (group S) , group I/R and ruthenium red (mitochondrial calcium uniporter inhibitor) group (group RR).Lung I/R was produced by clamping the left pulmonary hilum for 45 min followed by 2 h reperfusion in I/R and RR groups.In addition, ruthenium red 2.5 mg/kg was injected via the tail vein at 1 h before occlusion of the left pulmonary hilum in RR group.Arterial blood samples were obtained at 2 h of reperfusion for blood gas analysis, and oxygenation index (PaO2/FiO2) was calculated.The rats were sacrificed, and then the lungs were removed for determination of wet/dry lung weight ratio (W/D ratio).Results Compared with group S, oxygenation index was significantly decreased, and W/D ratio was increased in I/R and RR groups.Oxygenation index was significantly higher, and W/D ratio was lower in group RR than in group I/R.Conclusion The mechanism underlying lung I/R injury is related to the opening of mitochondrial calcium uniporter in rats.
