目的:探讨新型基因重组藻胆蛋白对小鼠S180实体瘤突变型P53表达的影响。方法将小鼠腋下接种S180细胞荷瘤造模后,随机分为阴性组,环磷酰胺组,新型基因重组藻胆蛋白组[50 mg/(kg·d)],天然藻蓝蛋白低、高浓度组[25,100 mg/(kg·d)],共5组,阴性组给予等量生理盐水,静脉注射给药11 d后处死,剥离肿瘤,采用免疫组化方法检测肿瘤组织中突变型P53的表达。结果新型基因重组藻胆蛋白组,天然藻蓝蛋白低、高浓度组突变型P53平均阳性表达率分别为12.28%,18.76%,15.80%,均低于阴性组的60.08%(P<0.05)。结论新型基因重组藻胆蛋白可明显抑制突变型P53表达(P<0.05),可能对突变型P53表达有下调作用。

목적:탐토신형기인중조조담단백대소서S180실체류돌변형P53표체적영향。방법장소서액하접충S180세포하류조모후,수궤분위음성조,배린선알조,신형기인중조조담단백조[50 mg/(kg·d)],천연조람단백저、고농도조[25,100 mg/(kg·d)],공5조,음성조급여등량생리염수,정맥주사급약11 d후처사,박리종류,채용면역조화방법검측종류조직중돌변형P53적표체。결과신형기인중조조담단백조,천연조람단백저、고농도조돌변형P53평균양성표체솔분별위12.28%,18.76%,15.80%,균저우음성조적60.08%(P<0.05)。결론신형기인중조조담단백가명현억제돌변형P53표체(P<0.05),가능대돌변형P53표체유하조작용。
Objective To explore the effect of new recombinant phycobiliprotein on the expression of P53 in mutant of mice treated with S180 solid tumor.Methods The tumor-bearing mice which were inoculated with S180 solid tumor were randomly divided into 5 groups:model group,cyclophosphamide group,new recombinant phycobiliprotein group [50 mg/(kg · d )],low,high dose of phycocyanin group[25,100 mg/(kg·d)],the same amount of sodium chloride injection was given to model group.After the intragastric administra-tion for 1 1 d,the mice were put to death and the solid tumor was removed from each group,and the expression of P53 mutant was detected by immunohistochemical SABC method.Results The average positive expression rates of P53 mutant among new recombinant phycobiliprotein group,low,high dose of phycocyanin group were 12.28%,18.76%,15.80% respectively,which were much lower than 60.08% of the model group(P <0.05).Conclusion New recombinant phycobiliprotein may decrease the expression of P53 mutant significantly,and may have down-regulatory effect on the expression of P53 mutant.
