目的:建立耐多西紫杉醇的人非小细胞肺癌细胞株 A549/ Docetaxel,并初步探索其与上皮间质转化(EMT)的关系。方法:用抗肿瘤药物多西紫杉醇体外以浓度递增和短时反复作用的方法诱导和筛选人非小细胞肺癌 A549细胞耐多西紫杉醇的细胞株;用 MTT 的方法检测细胞耐药指数及生长曲线,流式细胞仪检测细胞周期,RT - PCR 和 Western blot 检测 EMT 相关蛋白 fibronectin、vimentin 和 E - cadherin 的表达。结果:A549/ Docetaxel 对多西紫杉醇和紫杉醇的耐药指数分别为8.91和2.82;耐药细胞生长缓慢,倍增时间延长,S期细胞增多,G2期减少。549/ Docetaxel 与亲本 A549细胞相比,上皮细胞标志分子 E - cadherin 的表达减少,而间质细胞标志分子 fibronectin 和 vimentin 的表达增加。结论:成功建立了一株耐多西紫杉醇的细胞株A549/ Docetaxel,耐药的发生可能与 EMT 相关。

목적:건립내다서자삼순적인비소세포폐암세포주 A549/ Docetaxel,병초보탐색기여상피간질전화(EMT)적관계。방법:용항종류약물다서자삼순체외이농도체증화단시반복작용적방법유도화사선인비소세포폐암 A549세포내다서자삼순적세포주;용 MTT 적방법검측세포내약지수급생장곡선,류식세포의검측세포주기,RT - PCR 화 Western blot 검측 EMT 상관단백 fibronectin、vimentin 화 E - cadherin 적표체。결과:A549/ Docetaxel 대다서자삼순화자삼순적내약지수분별위8.91화2.82;내약세포생장완만,배증시간연장,S기세포증다,G2기감소。549/ Docetaxel 여친본 A549세포상비,상피세포표지분자 E - cadherin 적표체감소,이간질세포표지분자 fibronectin 화 vimentin 적표체증가。결론:성공건립료일주내다서자삼순적세포주A549/ Docetaxel,내약적발생가능여 EMT 상관。
Objective:To establish a new docetaxel resistant human non - small lung cancer cell line(A549 / Do-cetaxel)and investigate its association with epithelial - mesenchymal transition(EMT). Methods:Docetaxel resistant human non - small lung cancer cell line(A549 / Docetaxel)was developed by intermittent exposure to gradually in-creasing concentration of docetaxel from the parent cell line A549 in vitro. The resistance index(RI)of A549 / Do-cetaxel to anticancer agents and its growth rate were evaluated by MTT assay. The distribution of its cell cycle was de-tected by flow cytometry. The expression of fibronectin,vimentin and E - cadherin were detected by RT - PCR and Western blot. Results:The RI of A549 / Docetaxel cells to docetaxel and paclitaxel were respectively 8. 91 and 2. 82. Comparing with the parent cells,the multiplication time of A549 / Docetaxel cells was prolonged. The proportion of cells in G2 - phase decreased while those in S - phase increased,and the morphology of the resistant cells was changed obviously. Furthermore,we found the expression of fibronectin and vimentin in resistant cells were significant-ly higher than that in their parent cells while E - cadherin was much less expressed. Conclusion:We established a typical docetaxel resistant human non - small lung cancer cell line(A549 / Docetaxel),and it was suggested that the drug resistance of A549 / Docetaxel was related to EMT.
