目的:研究胡黄连苷Ⅱ治疗大鼠脑缺血损伤的最佳剂量和时间窗。方法通过双侧颈总动脉结扎法(BCCAO)建立大鼠前脑缺血模型,采用正交试验设计分组,经由腹腔注射胡黄连苷Ⅱ干预治疗,应用黄嘌呤氧化酶法、化学比色法和光化学法分别检测血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)的活性。结果根据血清SOD,GSHPx和CAT活性分析,应用胡黄连苷Ⅱ治疗脑缺血损伤的最佳效果为脑缺血1.5 h腹腔注射20 mg/kg,1.5 h/10(mg·kg)和1.5 h/20(mg·kg)。结论胡黄连苷Ⅱ治疗脑缺血损伤的最佳治疗时间窗和剂量为脑缺血1.5~2.0 h和腹腔注射10~20 mg/kg。

목적:연구호황련감Ⅱ치료대서뇌결혈손상적최가제량화시간창。방법통과쌍측경총동맥결찰법(BCCAO)건립대서전뇌결혈모형,채용정교시험설계분조,경유복강주사호황련감Ⅱ간예치료,응용황표령양화매법、화학비색법화광화학법분별검측혈청초양화물기화매(SOD)、곡광감태과양화물매(GSH-Px)화과양화경매(CAT)적활성。결과근거혈청SOD,GSHPx화CAT활성분석,응용호황련감Ⅱ치료뇌결혈손상적최가효과위뇌결혈1.5 h복강주사20 mg/kg,1.5 h/10(mg·kg)화1.5 h/20(mg·kg)。결론호황련감Ⅱ치료뇌결혈손상적최가치료시간창화제량위뇌결혈1.5~2.0 h화복강주사10~20 mg/kg。
Objective To study the optimal dose and time window of Picroside II in treating cerebral ischemia injury in rats.Methods The bilateral common carotid arteryligation(BCCAO)was used to establish the model of forebrain ischemia in rats,the orthogonal exper-iment design was used to divide the groups.The rats were given intraperitoneal injection of Picroside Ⅱ.The serum superoxide dismutase (SOD),glutathione peroxidase(GSH-Px)and catalase(CAT)activity was detect by the application of xanthine oxidase,chemical colorime-try and photochemical method.Results According to the analysis of serum SOD,GSH-Px and CAT activity in cerebral ischemia,the optimal effect of Picroside Ⅱin treating cerebral ischemic injury was by intraperitoneal injection of 1.5 h 20 mg/kg,1.5 h/10 (mg·kg) and 1.5 h/20 (mg·kg).Conclusion The optimal dose and time window of Picroside Ⅱin treating cerebral ischemia is by intraperi-toneal injection of 1.5 -2.0 h and 10-20 mg/kg.
