目的:探讨3种机械敏感通道在 AMsh 神经胶质细胞对机械刺激钙信号响应中的作用,为医学研究奠定基础。方法运用分子遗传学构建一系列转基因线虫品系,通过在体胞内钙成像技术,检测并分析 AMsh 神经胶质细胞对机械刺激的钙信号变化。结果机械刺激可引起 AMsh 神经胶质细胞的钙浓度升高,野生型荧光比值(F GCamp5.0/F mCher y )升高,与 trp-4(sy695);deg-1(u38)双基因突变体荧光比值升高差异无统计学意义(P >0.05),与 trp-4(sy695);mec-4(e1611)双基因突变体荧光比值升高差异有统计学意义(P <0.05)。结论机械敏感通道 TRP-4和 DEG-1均不参与 AMsh 神经胶质细胞对机械刺激的响应,但是机械敏感通道 MEC-4参与了 AMsh 神经胶质细胞对机械刺激的响应。AMsh 神经胶质细胞上的机械敏感受体可能是某种全新的离子通道;ALM 等触觉神经元可能对 AMsh 神经胶质细胞的活性起着调节作用。

목적:탐토3충궤계민감통도재 AMsh 신경효질세포대궤계자격개신호향응중적작용,위의학연구전정기출。방법운용분자유전학구건일계렬전기인선충품계,통과재체포내개성상기술,검측병분석 AMsh 신경효질세포대궤계자격적개신호변화。결과궤계자격가인기 AMsh 신경효질세포적개농도승고,야생형형광비치(F GCamp5.0/F mCher y )승고,여 trp-4(sy695);deg-1(u38)쌍기인돌변체형광비치승고차이무통계학의의(P >0.05),여 trp-4(sy695);mec-4(e1611)쌍기인돌변체형광비치승고차이유통계학의의(P <0.05)。결론궤계민감통도 TRP-4화 DEG-1균불삼여 AMsh 신경효질세포대궤계자격적향응,단시궤계민감통도 MEC-4삼여료 AMsh 신경효질세포대궤계자격적향응。AMsh 신경효질세포상적궤계민감수체가능시모충전신적리자통도;ALM 등촉각신경원가능대 AMsh 신경효질세포적활성기착조절작용。
Objective To investigate effects of three mechano-sensitive channels on the response of AMsh glial cells′calcium signaling to mechanical stimulation lay the foundation of medical research.Methods A series of transgenic strains of C.elegans by molecular genetics were constructed,and the response of AMsh glial cells′calcium signaling to the mechanical stimulation was detected and analyzed by the calcium imaging technique in vivo.Results Mechanical stimulation can induce the increase of calcium concentration in AMsh cells and the trp-4 (sy695);deg-1 (u38)double mutation′s increased fluorescence ratio (FGCamp5.0/FmCherry )has no significant difference to wild type′s (P > 0.05 );However,the trp-4 (sy695 );mec-4 (e1611)double mutation′s increased fluorescence ratio has no significant difference (P <0.05).Conclu-sion Mechano-gated ion channel MEC-4,but not TRP-4 and DEG-1 is involved in tactile stimulation-evoked responses in the AMsh glial cells.Our results suggest that MEC-4-expressing touch receptor neu-rons may contribute to the tactile stimulation-evoked responses in the AMsh glial cells.Moreover,some novel mechano-gated channel (s)may be expressed in C.elegans as mechano-sensor(s).
