Tongfeng Xiaoting Decoction%acute gouty arthritis%inflammatory Factors
目的:研究痛风消停汤对急性痛风性关节炎(AGA)大鼠受试踝关节滑膜组织炎性因子的影响.方法:60只SD雄性大鼠随机分为正常组、模型组、秋水仙碱组,痛风消停汤低、中、高剂量组,每组10只.正常组常规喂养,其余各组采用尿酸钠(MSU)溶液注射法建立急性痛风性关节炎模型.造模前2日,正常组与模型组按20mL/kg予以生理盐水灌胃,秋水仙碱组0.3mg/kg灌胃,1日1次;痛风消停汤低、中、高剂量组分别按2g/kg、4g/kg、8g/kg体质量给予痛风消停汤灌胃,1日1次,直至造模后第7日,连续9日.实验结束后,检测血尿酸,采用ELISA法检测受试踝关节IL-1β、IL-6、TNF-α的含量,免疫组化法检测TGF-β及其受体(TGF-βR1)的表达.结果:与正常组比较,模型组大鼠血尿酸、受试踝关节滑膜组织中IL-1β、IL-6、TNF-α含量明显升高(P<0.05);与模型组比较,痛风消停汤各组大鼠血尿酸、受试踝关节滑膜组织中IL-1β、IL-6、TNF-α含量明显降低(P<0.05),而痛风消停汤各组TGF-β1及TGF-βR1的表达较其他各组明显增强(P<0.05).结论:痛风消停汤治疗急性痛风性关节炎的机制可能与上调TGF-β1及TGF-βR1的表达、抑制IL-1β、IL-6、TNF-α的表达有关.

목적:연구통풍소정탕대급성통풍성관절염(AGA)대서수시과관절활막조직염성인자적영향.방법:60지SD웅성대서수궤분위정상조、모형조、추수선감조,통풍소정탕저、중、고제량조,매조10지.정상조상규위양,기여각조채용뇨산납(MSU)용액주사법건립급성통풍성관절염모형.조모전2일,정상조여모형조안20mL/kg여이생리염수관위,추수선감조0.3mg/kg관위,1일1차;통풍소정탕저、중、고제량조분별안2g/kg、4g/kg、8g/kg체질량급여통풍소정탕관위,1일1차,직지조모후제7일,련속9일.실험결속후,검측혈뇨산,채용ELISA법검측수시과관절IL-1β、IL-6、TNF-α적함량,면역조화법검측TGF-β급기수체(TGF-βR1)적표체.결과:여정상조비교,모형조대서혈뇨산、수시과관절활막조직중IL-1β、IL-6、TNF-α함량명현승고(P<0.05);여모형조비교,통풍소정탕각조대서혈뇨산、수시과관절활막조직중IL-1β、IL-6、TNF-α함량명현강저(P<0.05),이통풍소정탕각조TGF-β1급TGF-βR1적표체교기타각조명현증강(P<0.05).결론:통풍소정탕치료급성통풍성관절염적궤제가능여상조TGF-β1급TGF-βR1적표체、억제IL-1β、IL-6、TNF-α적표체유관.
Objective:To investigate the effect of Tongfeng Xiaoting Decoction on the inflammatory factors in ankle joint synovial tissue of acute gouty arthritis rats(AGA).Method:60 male SD rats were randomly divided into 6 groups:normal group, model group, colchicine group and low, medium and high Tongfeng Xiaoting Decoction groups (10 in each group).AGA model was established by induced monosodium urate (MSU) method except the normal group. Two days before AGA model was established, rats in normal and model groups received intra-gastric normal saline(NS) by 20ml/kg.d for qd, colchicine group received colchicine by 0.3mg/kg.d for qd, while those in low, medium and high Tongfeng Xiaoting Decoction groups received Tongfeng Xiaoting Decoction at the liquid of 2g /kg,4g /kg and 8g/kg respectively, once daily, until 7 days after model was formed, continuously for 9 days. After the experiment, serum uric acid was detected, the expression of IL-1β、IL-6、TNF-α、TGF-β1 were detected by ELISA and the expression of TGF-β1 and TGF-βR1 were detected by immunohistochemistry. Result:Serum uric acid and the content of IL-1β、IL-6、TNF-α、TGF-β1 in ankle joint synovial membrane were increased significantly in the model group compared to that of the normal group(P<0.05).Serum uric acid and the content of IL-1β、IL-6、TNF-α、TGF-β1 in ankle joint synovial membrane were decreased significantly in Tongfeng Xiaoting Decoction groups compared to that of the model group(P<0.05). The expression of TGF-β1and TGF-βR1 in ankle joint synovial membrane were increased significantly in Tongfeng Xiaoting Decoction groups compared to that of the others group(P<0.05).Conclusion: The mechanism of treating gouty arthritis by Tongfeng Xiaoting Decoction may be through up-regulating the expression of TGF-β1 ,TGF-rβ1 and inhibiting the expression of IL-1β、IL-6、TNF-αin synovial tissue.
