目的:研究中药肾糖组方对糖尿病肾病大鼠C-Ⅳ、FN表达影响,探讨其在DN大鼠肾保护中的作用机制.方法:采用高脂饲料喂养+STZ腹腔注射制作DN大鼠模型. 分正常对照组、模型对照组、中药治疗组、西药治疗组(洛汀新组)、雷公藤治疗组、中西药治疗组(肾糖组方+洛汀新组) ,测定大鼠24 h尿蛋白定量,免疫组化方法测定大鼠肾组织中C-Ⅳ、FN表达. 结果:经中药肾糖组方治疗2个月后,24 h尿蛋白定量各治疗组均明显低于模型组(P<0. 05);大鼠模型组C-Ⅳ、FN分别呈现高阳性表达,明显高于各治疗组(P<0. 05). 结论:中药肾糖组方对DN大鼠有一定的治疗效果,可以改善肾功能,延缓糖尿病肾脏损伤,其可能机制是抑制了大鼠肾纤维化进展,与抑制了C-Ⅳ、FN在大鼠肾组织中表达有关.

목적:연구중약신당조방대당뇨병신병대서C-Ⅳ、FN표체영향,탐토기재DN대서신보호중적작용궤제.방법:채용고지사료위양+STZ복강주사제작DN대서모형. 분정상대조조、모형대조조、중약치료조、서약치료조(락정신조)、뢰공등치료조、중서약치료조(신당조방+락정신조) ,측정대서24 h뇨단백정량,면역조화방법측정대서신조직중C-Ⅳ、FN표체. 결과:경중약신당조방치료2개월후,24 h뇨단백정량각치료조균명현저우모형조(P<0. 05);대서모형조C-Ⅳ、FN분별정현고양성표체,명현고우각치료조(P<0. 05). 결론:중약신당조방대DN대서유일정적치료효과,가이개선신공능,연완당뇨병신장손상,기가능궤제시억제료대서신섬유화진전,여억제료C-Ⅳ、FN재대서신조직중표체유관.
Objective:To investigate the effect of Traditional Chinese medicine kidney sugar formula on expression of Fi-bronectin( FN) and typeⅣ collagen ( C-Ⅳ) of renal tissue in STZ-induced diabetic nephropathy rats. Methods:Fed with high fat diet+STZ produced by intraperitoneal injection in rats with diabetic nephropathy. Rats were randomly divided into five groups: nor-mal control group, model control group, Chinese medicine group ( Traditional Chinese medicine kidney sugar formula) , western medi-cine group ( Lotensin group) and Chinese and Western medicine group ( Traditional Chinese medicine kidney sugar formula+Lotensin group) . Rats were determined 24 h urinary protein and were measured by immunohistochemistry in rats C-Ⅳ, FN expression. Re-sults:After treatment of two months with Traditional Chinese medicine kidney sugar formula, 24 h urinary protein in the treatment groups were significantly lower than the model group (P< 0. 05);and C-IV and FN of Renal Tissue in the model rats showed high expression respectively, significantly higher than every treatment groups (P<0. 05). Conclusion:Renal sugar granule has a certain therapeutic effect on STZ-induced Diabetic Nephropathy Rats. Renal sugar granule can improve kidney function and delay diabetic kidney damage. The possible mechanism is the inhibition of the progress of renal fibrosis in rats, which was connected with the inhibi-ted expression of C-Ⅳ and FN in renal tissue in rats.
