目的::探讨异丙酚对严重烫伤大鼠早期肝脏的保护作用。方法:SD 大鼠40只,随机分为异丙酚保护组和对照组,每组各20只。制作大鼠30%体表面积Ⅲ度烫伤模型。分别于烫伤前(0 h)、烫伤后第6、12、24 h 取下腔静脉血和肝组织匀浆,ELISA 法测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,紫外可见分光光度计检测肝组织丙二醛(MDA)水平、超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)水平,同时取大鼠肝脏 HE 染色进行病理学观察。结果:烫伤后6、12、24 h 较0 h ALT、AST 和 MDA 显著增高,SOD 和 GSH 显著降低,异丙酚保护组可显著降低 ALT、AST 和 MDA 水平,提高 SOD 活性和GSH 水平,组内差异显著(P <0.05);组间差异显著(P <0.05)。结论:异丙酚对严重烫伤早期肝脏具有保护作用,与其抗肝脏缺血再灌注损伤作用和抑制自由基的生成和释放有关。

목적::탐토이병분대엄중탕상대서조기간장적보호작용。방법:SD 대서40지,수궤분위이병분보호조화대조조,매조각20지。제작대서30%체표면적Ⅲ도탕상모형。분별우탕상전(0 h)、탕상후제6、12、24 h 취하강정맥혈화간조직균장,ELISA 법측정혈청곡병전안매(ALT)、곡초전안매(AST)활성,자외가견분광광도계검측간조직병이철(MDA)수평、초양화물기화매(SOD)활성、곡광감태(GSH)수평,동시취대서간장 HE 염색진행병이학관찰。결과:탕상후6、12、24 h 교0 h ALT、AST 화 MDA 현저증고,SOD 화 GSH 현저강저,이병분보호조가현저강저 ALT、AST 화 MDA 수평,제고 SOD 활성화GSH 수평,조내차이현저(P <0.05);조간차이현저(P <0.05)。결론:이병분대엄중탕상조기간장구유보호작용,여기항간장결혈재관주손상작용화억제자유기적생성화석방유관。
Objective:To explore the protective effect of propofol to rats liver on early severe scald.Methods:A total of 40 SD adult rats were randomly divided into control group and propofol protection group,with 20 rats in each group.A model with Ⅲ degree (30% of total body surface area)scald injury was established.Respectively at before scald(0 h),and after scald 6,12,24 h postcava blood and liver tissues were taken.Serum ALT and AST were examined by ELISA.Liver MDA, SOD and GSH were examined by Ultraviolet spectrophotometer.Pathological changes of liver was observed in HE stain. Results:After 6,12,24 h scald,ALT,AST and MDA were markly increased,and SOD and GSH were obviously decreased than 0 h;Propofol protection group can significantly reduce the level of ALT,AST and MDA,improve the activity of SOD and GSH;the differences in the group were significant(P <0.05),and the differences between groups were significant(P <0.05).Conclusion:Liver protective of Propofol for severe scald early,and relevant with resisting hepatic ischemia-reperfusion injury and inhibit the formation of free radicals and release.
