目的:研究高蛋氨酸(Methionine,Met)饮食对大鼠血浆同型半胱氨酸(Homocysteine,Hcy)和硫腺苷同型半胱氨酸(S-adenosylhomocysteine,SAH)浓度及颈动脉血管平滑肌细胞的影响,同时观察叶酸、维生素 B12对其血管损害的保护作用。方法选取 Wistar 雄性大鼠21只,随机分为三组:对照组给予正常饲料,高蛋氨酸组给予添加1%高蛋氨酸的饲料;干扰组给予1%高蛋氨酸同时添加叶酸、维生素 B12饲料。饲喂动物10周后留取腹主动脉血检测Hcy、SAH 浓度,留取颈总动脉标本行 HE 染色,并计算平滑肌厚度指数(Smooth muscle thickness index,SMTI)。结果高蛋氨酸组大鼠血浆 Hcy 水平较对照组及干扰组明显增高(P <0.05),血浆 SAH 水平较对照组明显下降(P <0.05),SMTI 较对照组及干扰组明显增高(P <0.05)。结论高蛋氨酸饮食对 Wistar 雄性大鼠具有毒性作用,能引起血浆 Hcy 水平升高,SAH 水平下降,引起动脉平滑肌损伤,并且这种血管损伤可能是由于血浆 Hcy 水平增高所致。叶酸、维生素 B12具有拮抗高蛋氨酸饮食所致的高 Hcy 血症,对血管损伤具有一定的保护作用。

목적:연구고단안산(Methionine,Met)음식대대서혈장동형반광안산(Homocysteine,Hcy)화류선감동형반광안산(S-adenosylhomocysteine,SAH)농도급경동맥혈관평활기세포적영향,동시관찰협산、유생소 B12대기혈관손해적보호작용。방법선취 Wistar 웅성대서21지,수궤분위삼조:대조조급여정상사료,고단안산조급여첨가1%고단안산적사료;간우조급여1%고단안산동시첨가협산、유생소 B12사료。사위동물10주후류취복주동맥혈검측Hcy、SAH 농도,류취경총동맥표본행 HE 염색,병계산평활기후도지수(Smooth muscle thickness index,SMTI)。결과고단안산조대서혈장 Hcy 수평교대조조급간우조명현증고(P <0.05),혈장 SAH 수평교대조조명현하강(P <0.05),SMTI 교대조조급간우조명현증고(P <0.05)。결론고단안산음식대 Wistar 웅성대서구유독성작용,능인기혈장 Hcy 수평승고,SAH 수평하강,인기동맥평활기손상,병차저충혈관손상가능시유우혈장 Hcy 수평증고소치。협산、유생소 B12구유길항고단안산음식소치적고 Hcy 혈증,대혈관손상구유일정적보호작용。
Objective To investigate the impact of High methionine diet on the concentration of Homocysteine (Hcy)and S-adenosylhomocysteine(SAH)and the carotid artery vascular smooth muscle cells in rats,observe the pro-tective effect of folic acid and vitamin B12 on the vascular damage.Methods 21 male Wistar rats were selected,ran-domly divided into three groups:The control group received normal diet,the high methionine group were added 1%high methionine on the basis of normal diet,the intervention group adding folic acid and vitamin B12 on the basis of the high methionine group.Feeding these diets for 10 weeks,then collecting the abdominal aortic blood for detection the concentrations of Hcy and SAH,isolating the carotid artery for HE staining and calculating the smooth muscle thick-ness index (SMTI).Results The concentration of homocysteine in the high methionine group was significantly higher than the control group and the intervention group (P <0.05),the SAH level was significantly reduced contrary to the control group(P <0.05),SMTI was significantly higher than the control group and the intervention group (P <0.05). Conclusion High methionine diet has a toxic effect on the male Wistar rats,it can lead to the elevated of Hcy concen-tration,the decreased of plasma SAH level and the damaged of vascular smooth muscle cell,besides,the elevated homo-cysteine level may be the reason that causes this vascular damage.Folic acid,vitamin B12 have the effects that antago-nist the hyperhomocystinemia which are leaded by the high methionine diet and has a protective effect on the vascular damage.
