目的:探讨慢病毒介导碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)基因修饰的骨髓基质细胞(bone marrow stromal cells,BMSCs)对脑梗死后动脉生成的影响。方法利用慢病毒载体介导 bFGF 基因修饰 BMSCs,获得稳定转染 bFGF 的 BMSCs。脑梗死后1 d 立体定向移植至梗死灶周围。术后14 d 后采用免疫组织化学染色法标记梗死灶周围微小动脉,并采用 MCID 计算机成像分析系统测量微小动脉的密度及直径。结果与对照组比较,BMSCs 组及 bFGF-BMSCs 组微小动脉密度及直径明显增加(P<0.05),且 bFGF-BMSCs 组微小动脉密度及直径显著高于 BMSCs 组(P <0.05)。结论bFGF 修饰的 BM-SCs 可有效促进脑梗死后动脉生成。

목적:탐토만병독개도감성성섬유세포생장인자(basic fibroblast growth factor,bFGF)기인수식적골수기질세포(bone marrow stromal cells,BMSCs)대뇌경사후동맥생성적영향。방법이용만병독재체개도 bFGF 기인수식 BMSCs,획득은정전염 bFGF 적 BMSCs。뇌경사후1 d 입체정향이식지경사조주위。술후14 d 후채용면역조직화학염색법표기경사조주위미소동맥,병채용 MCID 계산궤성상분석계통측량미소동맥적밀도급직경。결과여대조조비교,BMSCs 조급 bFGF-BMSCs 조미소동맥밀도급직경명현증가(P<0.05),차 bFGF-BMSCs 조미소동맥밀도급직경현저고우 BMSCs 조(P <0.05)。결론bFGF 수식적 BM-SCs 가유효촉진뇌경사후동맥생성。
Objective To investigate the influence of lentiviral vector mediated basic fibroblast growth factor (bFGF)gene modified bone marrow stromal cells (BMSCs)on arteriogenesis in rats with cerebral ische-mia.Methods Rat BMSCs were lentivirally tranduced to express bFGF stably.They were transplanted into the ischemic boundary zone 1 d after middle cerebral artery occlusion.The arteriole were labeled by immunohis-tochemistry staining 14 days after MCAO and the arteriolar density and diameter were measured in the ischemic boundary(IBZ)using the MCID computer imaging analysis system.Results Compared with control group, treatment with BMSCs and bFGF-BMSCs significantly increased the arteriolar density and diameter in the IBZ (P < 0.05 ).The arteriolar density and diameter in bFGF-BMSCs group significantly increased than that in BMSCs group (P <0.05).Conclusions bFGF gene-modified BMSCs can efficiently improve arteriogenesis af-ter stroke.
