目的 探讨金松双黄酮联用紫杉醇对 A549 细胞 caspase -3 蛋白及Bcl-2、Bax mRNA 表达的作用. 方法 实验分为对照组、紫杉醇组(浓度1 μg? L-1)、金松双黄酮+紫杉醇组(浓度分别为0.5,1.0 μg? L-1),在药物作用72 h后,用相差显微镜观察A549细胞形态学变化;逆转录-聚合酶链反应检测细胞中Bcl-2、Bax mRNA 水平;蛋白免疫印迹法检测caspase -3 蛋白的表达. 结果 与紫杉醇组相比,联用金松双黄酮组较多细胞出现细胞核浓缩、碎裂的凋亡特征,且caspase-3蛋白及Bax mRNA表达上调而Bcl-2mRNA表达下降. 结论 联用金松双黄酮可增强紫杉醇对A549 细胞凋亡作用,可能与调节Bc1-2、Bax及caspase-3的表达水平有关.

L-1),在藥物作用72 h後,用相差顯微鏡觀察A549細胞形態學變化;逆轉錄-聚閤酶鏈反應檢測細胞中Bcl-2、Bax mRNA 水平;蛋白免疫印跡法檢測caspase -3 蛋白的錶達. 結果 與紫杉醇組相比,聯用金鬆雙黃酮組較多細胞齣現細胞覈濃縮、碎裂的凋亡特徵,且caspase-3蛋白及Bax mRNA錶達上調而Bcl-2mRNA錶達下降. 結論 聯用金鬆雙黃酮可增彊紫杉醇對A549 細胞凋亡作用,可能與調節Bc1-2、Bax及caspase-3的錶達水平有關.
목적 탐토금송쌍황동련용자삼순대 A549 세포 caspase -3 단백급Bcl-2、Bax mRNA 표체적작용. 방법 실험분위대조조、자삼순조(농도1 μg? L-1)、금송쌍황동+자삼순조(농도분별위0.5,1.0 μg? L-1),재약물작용72 h후,용상차현미경관찰A549세포형태학변화;역전록-취합매련반응검측세포중Bcl-2、Bax mRNA 수평;단백면역인적법검측caspase -3 단백적표체. 결과 여자삼순조상비,련용금송쌍황동조교다세포출현세포핵농축、쇄렬적조망특정,차caspase-3단백급Bax mRNA표체상조이Bcl-2mRNA표체하강. 결론 련용금송쌍황동가증강자삼순대A549 세포조망작용,가능여조절Bc1-2、Bax급caspase-3적표체수평유관.
Objective To discuss the influence of Sciadopitysin com-bined with paclitaxel on the expression of caspase -3 protein and Bcl-2, Bax genes in A549 cells.Methods The experiment was divi-ded into three groups:control , paclitaxel and sciadopitysin +paclitaxel groups.After treatment 72 h, the morphologic change was observed by phase contrast microscope .The mRNA expression of Bcl -2 and Bax was detected by reverse transcription -PCR.The expression of caspase -3 protein was analyzed by Western -blot.Results Compared with other groups , more A 549 cells treated by Sciadopitysin + paclitaxelare appeared apoptosis characteristics nuclear condensation and fragmenta-tion.Compared with paclitaxel group , in Sciadopitysin +paclitaxelare group, the mRNA level of Bcl-2 was down-regulated, while the pro-tein level of caspase -3 and mRNA level of Bax were up -regulated. Conclusion Sciadopitysin+paclitaxelare strengthened the action of pa-clitaxel to apoptosis of A549 cells, which might be correlated with the regulation of expressions of Bcl -2 , Bax and caspase -3.
