本文利用环介导等温扩增技术(loop-mediated isothermal amplification,LAMP),针对西瓜嗜酸菌(Aci-dovorax citrulli )基因组中 Aave_4063和 Aave_4064序列设计了2对特异引物 Ac-F3/Ac-B3和 Ac-FIP /Ac-BIP,建立了西瓜嗜酸菌的 LAMP 检测体系。利用该体系在65℃保温1 h,通过荧光显色即可完成检测。设计的引物特异性强,其检测灵敏度为2.0×101 cfu/mL。该方法为西瓜嗜酸菌的检疫及其所致病害的快速诊断提供了新的技术。
本文利用環介導等溫擴增技術(loop-mediated isothermal amplification,LAMP),針對西瓜嗜痠菌(Aci-dovorax citrulli )基因組中 Aave_4063和 Aave_4064序列設計瞭2對特異引物 Ac-F3/Ac-B3和 Ac-FIP /Ac-BIP,建立瞭西瓜嗜痠菌的 LAMP 檢測體繫。利用該體繫在65℃保溫1 h,通過熒光顯色即可完成檢測。設計的引物特異性彊,其檢測靈敏度為2.0×101 cfu/mL。該方法為西瓜嗜痠菌的檢疫及其所緻病害的快速診斷提供瞭新的技術。

The loop-mediated isothermal amplification (LAMP)was established for rapid detection of Acidovorax citrul-li .Four primers,Ac-F3/Ac-B3 and Ac-FIP/Ac-BIP,were designed specifically to recognize the Aave _4063 and Aave _4064 genes of A .citrulli .The LAMP products was obtained under isothermal conditions at 65 ℃ for 1 h.The result showed that no amplicon was observed except A .citrulli strains.The detection limits of pure cultures were 2.0×10 1 cfu/mL.This LAMP detection system provides a new alternative method for the rapid detection of A .citrulli .
