[目的]研究祛疤汤及外源性TGF-β1对人体皮肤成纤维细胞增殖和Ki-67表达的影响,进一步探讨祛疤汤在皮肤创面修复过程中的可能机制。[方法]MTT法观察0、5、10、25、50、100 ng·mL-1的TGF-β1对成纤维细胞的增殖效果,选取最佳浓度;根据最佳浓度将成纤维细胞分为1640培养基组、25ng·mL-1 TGF-β1组、50mg·mL-1祛疤汤组、25ng·mL-1 TGF-β1+50mg·mL-1祛疤汤组,用MTT法检测各组对成纤维细胞增殖的影响。用Western blot法检测各组药物作用下成纤维细胞内增殖相关抗原Ki-67表达的影响。[结果]TGF-β1呈浓度依赖性促进成纤维细胞增殖,在25 ng· mL-1时促成纤维细胞增殖效应达到最高峰(P<0.05),祛疤汤可抑制TGF-β1的促成纤维细胞增殖作用(P<0.05);TGF-β1促进成纤维细胞内增殖相关抗原Ki-67的表达,祛疤汤可抑制TGF-β1促成纤维细胞内增殖相关抗原Ki-67的表达。[结论]祛疤汤能有效抑制成纤维细胞增殖,可能与祛疤汤抑制TGF-β1促成纤维细胞内Ki-67的表达有关。

[목적]연구거파탕급외원성TGF-β1대인체피부성섬유세포증식화Ki-67표체적영향,진일보탐토거파탕재피부창면수복과정중적가능궤제。[방법]MTT법관찰0、5、10、25、50、100 ng·mL-1적TGF-β1대성섬유세포적증식효과,선취최가농도;근거최가농도장성섬유세포분위1640배양기조、25ng·mL-1 TGF-β1조、50mg·mL-1거파탕조、25ng·mL-1 TGF-β1+50mg·mL-1거파탕조,용MTT법검측각조대성섬유세포증식적영향。용Western blot법검측각조약물작용하성섬유세포내증식상관항원Ki-67표체적영향。[결과]TGF-β1정농도의뢰성촉진성섬유세포증식,재25 ng· mL-1시촉성섬유세포증식효응체도최고봉(P<0.05),거파탕가억제TGF-β1적촉성섬유세포증식작용(P<0.05);TGF-β1촉진성섬유세포내증식상관항원Ki-67적표체,거파탕가억제TGF-β1촉성섬유세포내증식상관항원Ki-67적표체。[결론]거파탕능유효억제성섬유세포증식,가능여거파탕억제TGF-β1촉성섬유세포내Ki-67적표체유관。
Objective]To investigate the value of human skin fibroblasts and the ability to secrete collagen through the Dispel-scar decoction and exogenous TGF-β1, to further explore the possible mechanism of Dispel-scar decoction in the skin wound healing process.[Methods]Observe the fibroblast proliferation level under the action of different concentration of TGF-β1(0, 5, 10, 25, 50, 100ng·mL-1) by MTT;Observe the fibroblast proliferation level under the action of 1640 media group. Optimal concentration Dispel scar decoction group, Dispel scar decoction group, Optimal concentration Dispel-scar decoction combined with Dispel-scar decoction group by MTT. According to the results of MTT, detected the fibroblast proliferation of 25ng·mL-1 TGF-β1 and the expression of fibroblast proliferation associated antigen Ki-67 and collagen molecules by Dispel-scar decoction combined with 25ng·mL-1 TGF-β1. [Results]MTT results showed that the promoting fibroblast proliferation rate reached a peak by the 25ng·mL-1 TGF-β1(P<0.05), Dispel-scar decoction inhibited fibroblast cell proliferation by the 25ng·mL-1 TGF-β1; the comparison of the results from each group showed that TGF-β1 promoted the expression of fibroblast cell proliferation associated antigen Ki-67 and collagen molecules.Dispel-scar decoction inhibited the expression of fibroblast cell proliferation associated antigen Ki-67 and collagen molecules by the TGF-β1. [Conclusion]Dispel-scar decoction can effectively inhibit fibroblast cell proliferation,having something to do with Dispel-scar decoction inhibiting the expression of fibroblast cell proliferation associated antigen Ki-67 and collagen molecules.
