目的:建立逆转录聚合酶链反应(RT -PCR)检测乳腺癌核内原癌基因(nuclear oncogene,MYC)和黏蛋白1(Mucin1,MUC1)基因表达水平,探讨其联合检测在乳腺癌诊断和治疗监测中的应用。方法:建立 RT -PCR 反应,并以β2-微球蛋白为内对照测定30例健康女性体检者、52例良性乳腺肿瘤和103例乳腺癌外周血中 MYC 和 MUC1的表达量。结果:外周血中 MYC 和 MUC1在乳腺癌组表达率显著高于良性乳腺疾病组及健康体检组,而良性乳腺疾病组和健康体检组的表达率无差异。MUC1和 MYC 联合检测的特异性显著高于单一基因。结论:RT -PCR 联合检测外周血 MUC1和 MYC 基因表达,可提高对乳腺癌诊断的特异性,具有标本来源容易、操作简便和价格便宜等优点,可有效辅助诊断乳腺癌。

목적:건립역전록취합매련반응(RT -PCR)검측유선암핵내원암기인(nuclear oncogene,MYC)화점단백1(Mucin1,MUC1)기인표체수평,탐토기연합검측재유선암진단화치료감측중적응용。방법:건립 RT -PCR 반응,병이β2-미구단백위내대조측정30례건강녀성체검자、52례량성유선종류화103례유선암외주혈중 MYC 화 MUC1적표체량。결과:외주혈중 MYC 화 MUC1재유선암조표체솔현저고우량성유선질병조급건강체검조,이량성유선질병조화건강체검조적표체솔무차이。MUC1화 MYC 연합검측적특이성현저고우단일기인。결론:RT -PCR 연합검측외주혈 MUC1화 MYC 기인표체,가제고대유선암진단적특이성,구유표본래원용역、조작간편화개격편의등우점,가유효보조진단유선암。
Objective:To evaluate combined detection of MYC and MUC1 gene in the diagnosis of breast cancer. Methods:We used RT -PCR to detect MYC and MUC1 mRNA express in clinical samples from 30 health women,52 patients with benign breast disease and 103 patients with breast cancer.β2 -microglobin was used as internal control. Results:No significant difference was found in the positive rate of MYC and MUC1 between normal controls and be-nign breast disease group(P >0.05).The positive rate of MYC and MUC1 in breast cancer group were higher than those in normal controls and benign breast disease group(P <0.05).The specificity of combined detection of MYC and MUC1 gene express was higher than that of single gene detection(P <0.05).Conclusion:Combined detection of MYC and MUC1 in peripheral blood by RT -PCR can improve the specificity of diagnosis of breast cancer,with spec-imens from easy,simple and cheap,which can effectively assist the diagnosis of breast cancer.
