目的:建立反相/弱阳离子交换混合模式色谱(C18/WCX)对10种β-内酰胺类抗生素(头孢羟氨苄、头孢替唑钠、头孢呋辛钠、头孢唑啉、头孢拉定、头孢西酮、头孢孟多酯钠、头孢噻吩、哌拉西林、氯唑西林钠)同时检测的方法。方法以C18/WCX为固定相,以乙腈:甲酸铵:水为流动相,流速为1 mL/min;温度为30℃;检测波长为254 nm,并以常规反相色谱分析柱Unitary C18为对照。结果结果表明,中性条件下优化流动相洗脱梯度,10种β-内酰胺类抗生素在C18/WCX具有较高的分离选择性。通过缓冲盐浓度和pH值的调控,发现C18/WCX与样品间分离机制由反相作用力与离子交换2种作用力共同作用。结论研究表明,混合模式色谱在β-内酰胺类抗生素的分离分析中具有较大的潜力。

Objective To establish a method for simultaneous determination of 10 kinds ofβ-lactam antibiotics (cefadroxil, ceftezole sodium, cefuroxime sodium, cefazolin, cephradine, cefazedone, cemandil sodium, cephalothin, pipracil and cloxacillin sodium) by reversed-phase liquid chromatography/weak cation exchange (RPLC/WCX, C18/WCX).Methods Chromatographic separation was achieved on a C18/WCX, using a mixture of acetonitrile, ammonium formate and water as mobile phase with a flow rate at 1 mL/min and 30℃, and the detector was set at 254 nm.Results Compared with the conventional reversed-phase chromatography Unitary C18, the selectivity of C18/WCX could be improved. It was found that the mechanism of samples separated on C18/WCX were hydrophobic interaction and ion exchange interaction by regulated by the salt concentration and pH value.Conclusion It showed that the mixed-mode chromatography would be a potential approach for the analysis and separation ofβ-lactam antibiotics.
