目的:观察肿瘤坏死因子相关凋亡诱导配体对非小细胞肺癌小鼠模型肿瘤细胞凋亡诱导的作用。方法:30只小鼠接种浓度为5×106/ml非小细胞肺癌细胞Tc-1。荷瘤小鼠随机分为6组(空白对照组和5种浓度rhTRAIL实验组)。空白对照组每天注射一次灭菌生理盐水,实验组每天注射浓度不同的rhTRAIL溶液,各组均连续注射15d。每2d测量小鼠肿瘤长短直径,计算肿瘤体积。取小鼠脾脏细胞培养,测量培养液上清TNF-α浓度。结果:rhTRAIL给药组小鼠肿瘤体积明显低于空白对照组( P﹤0.05),随着rhTRAIL浓度的增加,肿瘤体积相应减小;小鼠的Tc-1细胞凋亡率随着rhTRAIL浓度增高,其凋亡率逐渐升高;rhTRAIL给药组小鼠的TNF-α浓度均明显高于空白对照组,组间差异具有显著性( P﹤0.05)。结论:rhTRAIL能有效增强非小细胞肺癌小鼠体内的细胞因子分泌,诱导荷瘤小鼠的肿瘤细胞凋亡,表明rhTRAIL对非小细胞肺癌小鼠的肿瘤细胞具有一定的抑制作用。

목적:관찰종류배사인자상관조망유도배체대비소세포폐암소서모형종류세포조망유도적작용。방법:30지소서접충농도위5×106/ml비소세포폐암세포Tc-1。하류소서수궤분위6조(공백대조조화5충농도rhTRAIL실험조)。공백대조조매천주사일차멸균생리염수,실험조매천주사농도불동적rhTRAIL용액,각조균련속주사15d。매2d측량소서종류장단직경,계산종류체적。취소서비장세포배양,측량배양액상청TNF-α농도。결과:rhTRAIL급약조소서종류체적명현저우공백대조조( P﹤0.05),수착rhTRAIL농도적증가,종류체적상응감소;소서적Tc-1세포조망솔수착rhTRAIL농도증고,기조망솔축점승고;rhTRAIL급약조소서적TNF-α농도균명현고우공백대조조,조간차이구유현저성( P﹤0.05)。결론:rhTRAIL능유효증강비소세포폐암소서체내적세포인자분비,유도하류소서적종류세포조망,표명rhTRAIL대비소세포폐암소서적종류세포구유일정적억제작용。
Objective:To observe the effect of tumor necrosis factor-related apoptosis-inducing ligand on non-small cell lung cancer mouse model of tumor cell apoptosis. Methods:Thirty mice were inoculated with non-small cell lung cancer cells,seeding concentration 5 × 106 Tc-1 cells. 30 tumor-bearing mice were randomly divided into six groups( control group and five concentrations rhTRAIL positive experimental groups). The control group was in-jected with NS,the positive control group was injected with solution of different concentrations rhTRAIL every day, each group of continuous injection 15d,length of the diameter of the tumor,tumor volume was calculated. Mouse spleen cells were measured for TNF-αconcentrations and compared. Results:In five positive tumor volume was sig-nificantly lower than the control group(P﹤0. 05);in 5 positive control groups rhTRAIL concentration increased,the tumor volume was reduced accordingly;in Tc - five groups positive rate of apoptosis rhTRAIL concentration in-creased,the apoptosis rate increased gradually;in 5 positive groups TNF-α concentrations were significantly higher than the control group,the difference was significant(P﹤0. 05). Conclusion:rhTRAIL can effectively enhance the non-small cell lung cancer in mice secrete cytoKines,effectively induce tumor cell non-small cell lung cancer in mice apoptosis. rhTRAIL can inhibit non small cell lung cancer cells in mice with a certain extent.
