目的：检测S100A4蛋白在上皮性卵巢癌中的表达及其与化疗耐药的关系。方法：选择81例卵巢上皮性癌组织，分为耐药组40例，敏感组41例，应用免疫组织化学法检测S100A4在卵巢上皮性癌组织中的表达情况；MTT法测定卡铂对细胞的半数抑制浓度（ IC50），采用Western Blot检测卡铂化疗敏感的卵巢癌细胞株SKOV3及卡铂化疗耐药的卵巢癌细胞株SKOV3/CB中S100A4的表达情况。结果：S100A4分别在卵巢癌组织和卵巢癌细胞耐药组中的表达高于敏感组，与卵巢癌的化疗耐药有关（ P﹤0.05）。S100A4在胞浆/胞核或胞核表达耐药组中高于敏感组，有统计学意义。卡铂对 SKOV3组和 SKOV3/CB 组的 IC50分别为28.83μmol/L和69.11μmol/L，因此SKOV3/CB对卡铂的耐药指数（ RI）为2.4。结论：S100A4的表达与上皮性卵巢癌的化疗耐药密切相关，为卵巢癌患者个体化治疗提供依据。
목적：검측S100A4단백재상피성란소암중적표체급기여화료내약적관계。방법：선택81례란소상피성암조직，분위내약조40례，민감조41례，응용면역조직화학법검측S100A4재란소상피성암조직중적표체정황；MTT법측정잡박대세포적반수억제농도（ IC50），채용Western Blot검측잡박화료민감적란소암세포주SKOV3급잡박화료내약적란소암세포주SKOV3/CB중S100A4적표체정황。결과：S100A4분별재란소암조직화란소암세포내약조중적표체고우민감조，여란소암적화료내약유관（ P﹤0.05）。S100A4재포장/포핵혹포핵표체내약조중고우민감조，유통계학의의。잡박대 SKOV3조화 SKOV3/CB 조적 IC50분별위28.83μmol/L화69.11μmol/L，인차SKOV3/CB대잡박적내약지수（ RI）위2.4。결론：S100A4적표체여상피성란소암적화료내약밀절상관，위란소암환자개체화치료제공의거。
Objective:To explore the relationship between chemoresistance and S100A4 expression in epithelial o-varian carcinoma. Methods:The expression of S100A4 was detected by SP immunohistochemistry method in 81 epi-thelial ovarian carcinoma which were divided into resistant group(40 cases)and sensitive group(41 cases). Western Blot was used to identify S100A4 protein level respectively carboplatin sensitive SKOV3 cell line and carboplatin re-sistant SKOV3/CB cell line. 50% inhibitory concentration( IC50 )to carboplatin in SKOV3 and SKOV3/CB was de-tected with MTT method. Results:The expression rate of S100A4 in epithelial ovarian carcinoma carboplatin resistant was higher than that in epithelial ovarian carcinoma carboplatin sensitive(P﹤0. 05). The expression of S100A4 had correlation with chemotherapy resistance in epithelial ovarian carcinoma. Both cytoplasmic and nuclear expression or nuclear expression of S100A4 in epithelial ovarian carcinoma carboplatin resistant was significantly higher than that in than those in epithelial ovar-ian carcinoma carboplatin sensitive(P﹤0. 05). The IC50 was 28. 83μmol/L in SKOV3 cells. The IC50 was 69. 11μmol/L in SKOV3/CB cells. Multidrug resistance index of carboplatin resistant SKOV3/CB was 2. 4. Conclusion:The expression of S100A4 is closely associated with chemotherapy-resistance of epithelial o-varian carcinoma and provide certain evidence for indivi-dualized treatment in ovarian cancer patients.
