目的:应用新的方法建立家兔深静脉血栓模型,并观察其伴发炎症反应的特点。方法取新西兰白兔30只,随机分为对照组12只,血栓模型组18只。对照组家兔仅行开腹游离后腔静脉,模型组家兔后腔静脉行不完全结扎加暂时性阻断术建立静脉血栓模型。建模后3、7、14 d分别进行大体观察、多普勒超声、组织病理学检查,并进行2组比较;计数模型组家兔病变静脉局部炎症细胞数,测定其血浆炎性因子白细胞介素‐6(IL‐6)、单核细胞趋化蛋白‐1(MCP‐1)的含量。结果造血栓模型手术顺利,成功率100%;术后1只家兔因腹泻死亡,死亡率3%。大体观察、超声检查及病理检测均证实模型组家兔血栓形成确切、稳定;而对照组无明显血栓形成。模型组家兔病变部炎症细胞聚集明显,不同细胞的数量随时间而变化;血浆炎性因子IL‐6、MCP‐1明显增高。结论后腔静脉不全结扎联合暂时性阻断是一种新的方法,可建立稳定的深静脉血栓动物模型;静脉血栓伴发的炎症具有持续性、全身性和急性向慢性转变的特点。

목적:응용신적방법건립가토심정맥혈전모형,병관찰기반발염증반응적특점。방법취신서란백토30지,수궤분위대조조12지,혈전모형조18지。대조조가토부행개복유리후강정맥,모형조가토후강정맥행불완전결찰가잠시성조단술건립정맥혈전모형。건모후3、7、14 d분별진행대체관찰、다보륵초성、조직병이학검사,병진행2조비교;계수모형조가토병변정맥국부염증세포수,측정기혈장염성인자백세포개소‐6(IL‐6)、단핵세포추화단백‐1(MCP‐1)적함량。결과조혈전모형수술순리,성공솔100%;술후1지가토인복사사망,사망솔3%。대체관찰、초성검사급병리검측균증실모형조가토혈전형성학절、은정;이대조조무명현혈전형성。모형조가토병변부염증세포취집명현,불동세포적수량수시간이변화;혈장염성인자IL‐6、MCP‐1명현증고。결론후강정맥불전결찰연합잠시성조단시일충신적방법,가건립은정적심정맥혈전동물모형;정맥혈전반발적염증구유지속성、전신성화급성향만성전변적특점。
Objective To establish a rabbit model of deep vein thrombosis by a new method ,and to observe its characteristics of concomitant inflammatory response .Methods Thirty New Zealand white rabbits were ran‐domly divided into two groups .Sham surgery by exposing of vena cava was performed in control group .Inferior vena cava thrombosis model was established by temporary blocking blood flow in inferior vena cava and incomplete ligation .At 3 days ,7 days and 14 days after modeling ,general observation ,ultrasound and pathological examina‐tion were taken .To count the local inflammatory cells number of model group lesions ,and detect the contents of inflammatory cytokines IL‐6 and MCP‐1 in plasma .Results The success rate of operation was 100% ,one rabbit died due to diarrhea after surgery in modeling group ,the mortality was 3% .Precise and stable thrombosis were confirmed in model group by general observation ,ultrasound examination and pathological examination ;There was no obvious thrombosis in control group .In model group ,inflammatory cells gathered in the vein wall obvious‐ly ,and different cells number changed with time ;Plasma inflammatory cytokines such as IL‐6 ,MCP‐1 increased significantly .Conclusion By the new method which blocked blood flow temporarily and ligated the vein incom‐pletely ,a stable model of venous thrombosis is feasible in rabbits ;Inflammation of the vein thrombosis is continu‐ous ,systemic and from acute to chronic .
