本研究针对鲤科疱疹病毒2型(CyHV-2)的主要衣壳蛋白基因,设计了特异性引物和Taqman探针,建立了实时荧光PCR方法。其检测下限为83拷贝,标准曲线R2>0.998,且经比对,灵敏度与现行方法相当。选取了健康异育银鲫、金鱼、鲤鱼以及常见4种水生DNA病毒以及7种水生环境细菌进行特异性实验,结果均无交叉反应。重复性实验显示,2份独立的15次重复结果变异系数均小于2%,说明本方法重复性好。本方法能作为目前CyHV-2疫病检测方法的有效补充,对该病的防控具有重要的意义。
本研究針對鯉科皰疹病毒2型(CyHV-2)的主要衣殼蛋白基因,設計瞭特異性引物和Taqman探針,建立瞭實時熒光PCR方法。其檢測下限為83拷貝,標準麯線R2>0.998,且經比對,靈敏度與現行方法相噹。選取瞭健康異育銀鯽、金魚、鯉魚以及常見4種水生DNA病毒以及7種水生環境細菌進行特異性實驗,結果均無交扠反應。重複性實驗顯示,2份獨立的15次重複結果變異繫數均小于2%,說明本方法重複性好。本方法能作為目前CyHV-2疫病檢測方法的有效補充,對該病的防控具有重要的意義。
본연구침대리과포진병독2형(CyHV-2)적주요의각단백기인,설계료특이성인물화Taqman탐침,건립료실시형광PCR방법。기검측하한위83고패,표준곡선R2>0.998,차경비대,령민도여현행방법상당。선취료건강이육은즉、금어、리어이급상견4충수생DNA병독이급7충수생배경세균진행특이성실험,결과균무교차반응。중복성실험현시,2빈독립적15차중복결과변이계수균소우2%,설명본방법중복성호。본방법능작위목전CyHV-2역병검측방법적유효보충,대해병적방공구유중요적의의。
Primers and Taqman probe targeting major capsid protein of cyprinid herpesvirus 2(CyHV-2)were designed and a real-time PCR method was established. The lower limit of detection was 83 copies,and standard curve R2>0.998. Comparison experiments showed that the sensitivity of this method was of same level as existing method.Healthy Carassius auratus gibelio,Carassius auratus auratus,Cyprinus Carpio,4 aquatic viruses and 7 aquatic bacteria were included for specificity tests,no cross reaction was observed. Repeatability tests showed that both coefficients of varia-tion of independent result repeated 15 times were smaller than 2%,suggesting that the repeatability of this method was ideal. The established method would be supplementary to the existing CyHV-2 detection methods,and was of important significance for disease control.
