Gas chromatography%Coupled method of distillation-extraction%Ligustilide%Volatile oil of Ligusticum Chuanx-iong Hort
目的:对蒸馏原萃取耦合法进行进一步优化,用于提取川芎挥发油。方法将蒸馏法与溶剂萃取法耦合用于提取川芎挥发油,以藁本内酯回收率为指标优选混有萃取剂的川芎挥发油的分离、纯化过程。用气相色谱法检测藁本内酯含量,采用AT-5石英毛细管色谱柱(30 m×0.32 mm×0.25μm);FID检测器;进样口温度:250℃,检测器温度:250℃;程序升温。结果确定优化后的提取工艺,即蒸馏-萃取所得川芎挥发油的石蜡溶液经1 ml乙醇萃取1次,0.5 ml乙醇萃取5次,1.5倍量30%NaCl水溶液盐析等得到川芎挥发油。结论优化后的蒸馏原萃取耦合法工艺稳定,可以用于提取川芎挥发油。

목적:대증류원췌취우합법진행진일보우화,용우제취천궁휘발유。방법장증류법여용제췌취법우합용우제취천궁휘발유,이고본내지회수솔위지표우선혼유췌취제적천궁휘발유적분리、순화과정。용기상색보법검측고본내지함량,채용AT-5석영모세관색보주(30 m×0.32 mm×0.25μm);FID검측기;진양구온도:250℃,검측기온도:250℃;정서승온。결과학정우화후적제취공예,즉증류-췌취소득천궁휘발유적석사용액경1 ml을순췌취1차,0.5 ml을순췌취5차,1.5배량30%NaCl수용액염석등득도천궁휘발유。결론우화후적증류원췌취우합법공예은정,가이용우제취천궁휘발유。
Objective To further optimize coupled method of distillation-extraction for volatile oil of Ligusticum Chuanxiong Hort.. Methods Distillation and solvent extraction method were coupled to extract volatile oil of Ligusticum Chuanxiong Hort..Recovery rate of ligustilide was selected as an indicator to optimize the separation and purification of volatile oil of Ligusticum Chuanxiong Hort.,which was mixed with extraction agent.The content of ligustilide was deter-mined by gas chromatography.AT-5 quartz capillary at size of 30 mí0.32 mmí0.25 μm was selected as capillary-col-umn chromatography.The basic conditions were flame ionization detector (FID),temperature of injection port being 250℃, temperature of detector being 250℃,and temperature programming. Results The optimized extraction process was paraffin wax solution extracted once by 1 ml ethyl alcohol,and 5 times by 0.5 ml ethyl alcohol,and salted out by 1.5 times of 30% sodium chloride solution before obtaining volatile oil of Ligusticum Chuanxiong Hort.. Conclusion The opti-mized coupled method of distillation-extraction process is stable and can be applied to extract volatile oil of Ligusticum Chuanxiong Hort..
