该文通过观察不同时期豉曲的菌丝生长情况,测定霉菌菌落总数、酶活力的动态变化,确定最佳的前发酵生产工艺,并对最佳条件下不同时期豉曲的酶活进行API ZYM(酶谱鉴定)系统的半定量分析。结果表明:在培养温度15℃、接种量1%、含水量45%、培养时间64 h时最佳,此时菌丝生长旺盛,产生大量孢子,霉菌菌落总数为2.1×107 CFU/g,总蛋白酶活力为103.33 U/g,纤维素酶活力为1.87 U/g,且通过API ZYM系统检测出了碱性磷酸盐酶、白氨酸芳胺酶、酸性磷酸酶、萘酚–AS–BI–磷酸水解酶等酶。

Toobtain theoptimal process parametersof the pro–fermentation processing,the growthof mycelium,the total numberof moldcolony,proteinase activity andcellulase activity wereobserved and determined. Under theoptimal pro–fermentation processing,the API ZYM system was applicated to semi–quantiative analysisof enzymatic activity. The results show that:at 15℃,64 h,1% inoculum size and 45% moisturecontent,hyphagrew vigorously,a large numberof spores produced,the total numberof moldcolony was 2.1×107 CFU/g,total protease activity andcellulose activity reached 103.33 U/g and 1.87 U/g,respectively. Alkaline phosphatase,Leucine arylamidase,Acid phosphatase and Naphtol–AS–BI–phosphohydrilase were detected by API ZYM system.
