目的:探讨恒定型自然杀伤T细胞( iNKT)在自发性早产发动中的作用。方法选择自发性早产孕妇40例(早产组)和正常孕妇40例(对照组)。采用流式细胞术检测蜕膜组织中iNKT细胞和自然杀伤( NK)细胞的表达;采用RT-PCR法检测穿孔素(perforin)mRNA的表达。结果早产组蜕膜组织中iNKT细胞和NK细胞的百分比分别为(1.05±0.29)%和(24.21±9.32)%,均显著高于对照组iNKT细胞(0.48±0.17)%和NK细胞(14.97±6.05)%的百分比(t值分别为2.267和2.094,均P<0.05);经Spearman秩相关分析显示,早产组iNKT细胞和NK细胞呈明显的正相关(r=0.634,P<0.05);RT-PCR结果显示,早产组穿孔素mRNA的表达也显著高于对照组( t=2.086,P<0.05)。结论 iNKT细胞可能通过产生穿孔素并促进NK细胞活化,参与自发性早产的免疫发病机制。

Objective To investigate the role of invariant natural killer T-cells ( iNKT) cells in the launch of spontaneous preterm labor. Methods Totally 40 preterm pregnant women ( preterm group ) and 40 normal pregnant women ( control group ) were selected.Flow cytometry was used to detect iNKT cells and NK cells in decidual tissue, and RT-PCR was used to detect the expression of perforin mRNA. Results The percentages of iNKT cells and NK cells in decidual tissue in the preterm group were (1.05 ±0.29)% and (24.21 ± 9.32)%, respectively, which were significantly higher than those of iNKT cells (0.48 ±0.17)%and NK cells (14.97 ±6.05)%in the control group (t value was 2.267 and 2.094, respectively, both P<0.05).Spearman rank correlation analysis showed that iNKT cells was positively related with NK cells in the preterm group (r=0.634,P<0.05).RT-PCR results revealed that the expression of perforin mRNA in the preterm group was significantly higher than that in the control group (t=2.086, P<0.05).Conclusion INKT cells may take part in the immune pathogenesis of spontaneous preterm labor through the production of perforin and the promotion of NK cell activation.
