目的::建立稳定的白背三七悬浮细胞培养体系。方法:采用植物组织培养技术,探讨初始接种密度、装液量、初始pH值、激素种类及浓度4个因素对白背三七细胞悬浮培养的影响。结果:白背三七细胞悬浮培养的适宜条件为:B5基本培养基+0.5 mg/L NAA+2.0 mg/L 6-BA+30g/L蔗糖,pH 7.0,装液量为40%,初始接种密度为90g/L(鲜质量),弱光培养。在此优化条件下,细胞生长呈“S”形曲线;培养过程中,培养基的pH呈先下降后缓慢升高的变化趋势;细胞活力的变化稍滞后于生长曲线,逐渐降低。
目的::建立穩定的白揹三七懸浮細胞培養體繫。方法:採用植物組織培養技術,探討初始接種密度、裝液量、初始pH值、激素種類及濃度4箇因素對白揹三七細胞懸浮培養的影響。結果:白揹三七細胞懸浮培養的適宜條件為:B5基本培養基+0.5 mg/L NAA+2.0 mg/L 6-BA+30g/L蔗糖,pH 7.0,裝液量為40%,初始接種密度為90g/L(鮮質量),弱光培養。在此優化條件下,細胞生長呈“S”形麯線;培養過程中,培養基的pH呈先下降後緩慢升高的變化趨勢;細胞活力的變化稍滯後于生長麯線,逐漸降低。
목적::건립은정적백배삼칠현부세포배양체계。방법:채용식물조직배양기술,탐토초시접충밀도、장액량、초시pH치、격소충류급농도4개인소대백배삼칠세포현부배양적영향。결과:백배삼칠세포현부배양적괄의조건위:B5기본배양기+0.5 mg/L NAA+2.0 mg/L 6-BA+30g/L자당,pH 7.0,장액량위40%,초시접충밀도위90g/L(선질량),약광배양。재차우화조건하,세포생장정“S”형곡선;배양과정중,배양기적pH정선하강후완만승고적변화추세;세포활력적변화초체후우생장곡선,축점강저。
Objective:to establish stable suspension cell cultures of Gynura divaricata(L.)DC. Method: The effecting factors of initial cultural density, volume amount, initial pH value, hormone types and concentration were studied by plant tissue culture technology. Results: The most suitable conditions of cell suspension cultures were found to be B5 medium supplemented with 0.5 mg/L NAA+2.0 mg/L 6-BA+30g/L sucrose, pH 7.0, 40% volume amount, 90g/L(fresh weight) initial cultural density with weak light training. Suspension cell growth curves were like “S”. The pH value cultured mediums were reduced at first, and then increased gradually. The change of cell vitality was reduced gradually lagging that of cell growth curves.
