目的:探讨GP6基因rs1654419位点多态性与石家庄市区及周边地区汉族人群下肢深静脉血栓形成的关系。方法应用DNA直接测序技术检测了石家庄市区及周边地区汉族人群中243例下肢深静脉血栓形成( DVT)患者和445例健康体检者的GP6基因rs1654419位点的多态性。同时检测2组的体重指数( BMI)、血浆D-二聚体( D-Dimer)、纤维蛋白原(Fbg)水平。结果病例组患者BMI、D-Dimer、Fbg水平明显高于对照组( P <0殚.05)。病例组患者AA、AG 和GG基因型的频率分别为16.9%、27.6%和55.5%,而正常对照组AA、AG、GG 基因型的频率分别为9.4%、20.9%和69.7%。深静脉血栓形成患者A等位基因频率为0.307,正常对照组的A等位基因频率为0.199,病例组明显高于对照组( P <0.05)。病例组和对照组中,按GG、AG、AA不同基因型,Fbg、D-Dimer水平依次升高( P <0.05)。 Logistic回归分析结果显示AA基因、A等位基因与下肢深静脉血栓形成的独立危险因素,OR(95%CI)值为2.951(1.198~4.687)( P <0.05)。结论石家庄市区及周边地区汉族人群中GP6基因rs1654419位点多态性与下肢深静脉血栓患者血栓形成有关。

Objective To investigate the correlation between single nucleotide polymorphism (SNPs) in rs1654419 locus of GP6 gene and lower limbs deep venous thrombosis ( DVT ) in Chinese Han population around Shijiazhuang city. Methods The genotype and allele frequencies of SNPs in rs1654419 locus of GP6 gene in downtown and periphery area of Shijiazhuang city were detected by DNA direct sequencing technology in 243 cases of lower limbs DVT ( case group) and 445 cases of healthy subjects ( control group) , at the same time, the body mass index ( BMI) , plasma levels of D-Dimer and Fbg were detected for both groups.Results The BMI, plasma levels of D-Dimer and Fbg in case group were significantly higher than those in control group ( P <0.05).The gene frequencies of AA, AG and GG genotypes in case group were 16.9%, 27.6%, 55.5%,respectively,however,which in control group were 9.4%,20.9%, 69.7%,respectively.The A allele frequency (0.307) in case group was significantly higher than that (0.199) in control group ( P <0.05).According to different genotype of GG, AG, AA, the levels of Fbg and D-Dimer in both groups were increased in order ( P <0.05 ) . Logistic regression analysis results showed that AA genotype and A allele were the independent risk factor of lower limbs DVT, with OR (95%CI) being 2.951 (1.198~4.687) ( P <0.05).Conclusion The SNPs in rs1654419 locus of GP6 gene are correlated to thrombosis in patients with lower limbs DVT of Chinese Han population in downtown and periphery area of Shijiazhuang city.
