目的:研究阿尔茨海默病( AD)患者外周血单核细胞CD33的表达情况,探讨其作为血液生物标志物的可行性。方法收集2011年1月至2013年3月徐州市第一人民医院收治的100例AD患者( AD组)和100例认知正常老年人(对照组)的临床资料和外周静脉血标本,分离外周血单核细胞,采用实时荧光定量多聚酶链反应测定CD33 mRNA表达水平,流式细胞仪分析CD33阳性细胞率。采用Pearson相关分析CD33表达与简易精神状态检查( MMSE)评分的关系。结果 AD患者CD33 mRNA表达水平和CD33阳性细胞率均显著低于对照组[0.89±0.28比1.06±0.33;(40±15)%比(55±21)%],差异有统计学意义(P<0.01)。 CD33 mRNA表达水平、CD33阳性细胞率与MMSE评分呈正相关(r=0.78,0.22,P<0.05)。结论 CD33可能参与AD的发病过程,CD33作为AD临床诊断生物标志物具有一定的可行性。

Objective To investigate the feasibility of CD33 expression in peripheral blood mononuclear cells( PBMCs) as a biomarker in Alzheimer disease( AD) . Methods A total of 200 cases in Xuzhou First People′s Hospital were selected from Jan. 2011 to Mar. 2013,including 100 AD patients and 100 controls. CD33 mRNA level was determined by real-time quantitative polymerase chain reaction and the CD33 positive monocytes was analyzed by flow cytometry. Pearson correlation was performed to analyze the correlation between CD33 expression and Mini Mental State Examination(MMSE) score. Results The expression of CD33 mRNA and proportion of CD33 positive monocytes were significantly decreased in AD patients compared withthecontrols[0.89±0.28vs1.06±0.33,(40±15)% vs(55±21)%,P<0.01].Regressionanaly-sis showed the expression of CD33 mRNA,CD33 positive monocytes had positive relationship with the MMSE score(r=0. 78,0. 22,P<0. 05). Conclusion CD33 may play an important role in the pathogenesis of AD and is feasible to serve as a blood biomarker in AD diagnosis.
