目的:研究沙利霉素(Sal)增强肝癌细胞对5‐氟尿嘧啶(5‐FU)敏感性及其机制,为抗药性原发性肝癌患者提供新的治疗方案。方法选用肝癌细胞株 HepG2、SMMC‐7721和 M HCC‐97H 为研究对象,运用 M TT 方法,克隆形成实验,流式细胞仪检测 Sal 和5‐FU 联合作用下对肝癌细胞增殖抑制率、克隆形成、细胞凋亡和肿瘤干细胞增殖的影响;Western‐blot 方法检测 Sal和5‐FU 联合作用对 Wnt/β‐catenin 信号通路的影响。结果 Sal 和5‐FU 联合作用能够显著抑制肝癌细胞增殖和克隆形成并诱导细胞凋亡,具有协同作用;5‐FU 能促进肝癌肿瘤干细胞增殖,而 Sal 降低5‐FU 的这一作用;同时 Sal 能抑制 Wnt/β‐catenin 信号通路。结论 Sal 通过抑制 Wnt/β‐catenin 信号通路增强肝癌细胞对5‐FU 的敏感性;Sal 和5‐FU 联合作用为抗药性原发性肝癌患者提供新的治疗方案。
目的:研究沙利黴素(Sal)增彊肝癌細胞對5‐氟尿嘧啶(5‐FU)敏感性及其機製,為抗藥性原髮性肝癌患者提供新的治療方案。方法選用肝癌細胞株 HepG2、SMMC‐7721和 M HCC‐97H 為研究對象,運用 M TT 方法,剋隆形成實驗,流式細胞儀檢測 Sal 和5‐FU 聯閤作用下對肝癌細胞增殖抑製率、剋隆形成、細胞凋亡和腫瘤榦細胞增殖的影響;Western‐blot 方法檢測 Sal和5‐FU 聯閤作用對 Wnt/β‐catenin 信號通路的影響。結果 Sal 和5‐FU 聯閤作用能夠顯著抑製肝癌細胞增殖和剋隆形成併誘導細胞凋亡,具有協同作用;5‐FU 能促進肝癌腫瘤榦細胞增殖,而 Sal 降低5‐FU 的這一作用;同時 Sal 能抑製 Wnt/β‐catenin 信號通路。結論 Sal 通過抑製 Wnt/β‐catenin 信號通路增彊肝癌細胞對5‐FU 的敏感性;Sal 和5‐FU 聯閤作用為抗藥性原髮性肝癌患者提供新的治療方案。
목적:연구사리매소(Sal)증강간암세포대5‐불뇨밀정(5‐FU)민감성급기궤제,위항약성원발성간암환자제공신적치료방안。방법선용간암세포주 HepG2、SMMC‐7721화 M HCC‐97H 위연구대상,운용 M TT 방법,극륭형성실험,류식세포의검측 Sal 화5‐FU 연합작용하대간암세포증식억제솔、극륭형성、세포조망화종류간세포증식적영향;Western‐blot 방법검측 Sal화5‐FU 연합작용대 Wnt/β‐catenin 신호통로적영향。결과 Sal 화5‐FU 연합작용능구현저억제간암세포증식화극륭형성병유도세포조망,구유협동작용;5‐FU 능촉진간암종류간세포증식,이 Sal 강저5‐FU 적저일작용;동시 Sal 능억제 Wnt/β‐catenin 신호통로。결론 Sal 통과억제 Wnt/β‐catenin 신호통로증강간암세포대5‐FU 적민감성;Sal 화5‐FU 연합작용위항약성원발성간암환자제공신적치료방안。
Objective To study salinomycin (Sal) enhance the sensitivity of liver cancer cell to 5‐fluorouracil (5‐FU) and its mechanism ,and to provide drug‐resistant primary hepatocellular carcinoma (HCC) patients with a new treatment .Methods Hepa‐toma cell line HepG2 ,SMMC‐7721 ,M HCC‐97H were used for the research .The effect of Sal combined with 5‐FU on the cell pro‐liferation inhibition rate ,colony formation ,apoptosis and tumor stem cell proliferation were detected by the M TT assay ,colony for‐mation assay ,flow cytometry .And the effect on Wnt/‐catenin when Sal combined with 5‐FU were detected by Western‐blot . Results Sal combined with 5‐FU significantly inhibited the liver cancer cell proliferation and colony formation and induced cell ap‐optosis ,showed the synergistic effect .5‐FU promoted the proliferation of hepatocarcinoma stem ,but Sal reduced the function of 5‐FU .At the same time Sal could inhibit the Wnt/‐catenin signal pathway .Conclusion The Sal can increase the sensitivity of hepato‐cellular carcinoma cells to 5‐FU by inhibiting Wnt/‐catenin signal pathway ;Sal combined with 5‐FU could provide drug‐resistant HCC patients with a new treatment .
