目的探讨促红细胞生成素对血管平滑肌细胞增殖和凋亡的作用及机制.方法噻唑蓝(MTT)实验和5-乙炔基-2’脱氧尿嘧啶核苷(EdU)实验检测促红细胞生成素对平滑肌细胞增殖的作用;原位缺口末端标记法(TUNEL)染色检测促红细胞生成素对平滑肌细胞凋亡的作用.结果 MTT结果显示促红细胞生成素可抑制平滑肌细胞增殖,使吸光度下降55％,细胞外信号调节激酶(ERK)信号通路的抑制则可部分反转促红细胞生成素对平滑肌细胞增殖的抑制作用,使其吸光度上升42.4％ (P ＜0.05);EdU增殖实验结果显示空白对照组平滑肌细胞增殖的比例为38.3％,促红细胞生成素可显著抑制增殖,使其增殖率下降49.6％,ERK信号通路的抑制则使促红细胞生成素组平滑肌细胞的增殖率升高了44.6％(P＜0.05);TUNEL染色结果显示促红细胞生成素可以显著促进平滑肌细胞凋亡,使其凋亡的比例升高3.14倍(P＜0.05),ERK信号通路的阻断对平滑肌细胞的凋亡无显著影响(P＞0.05).结论促红细胞生成素能有效抑制血管平滑肌细胞增殖和促进其凋亡,进而抑制血液透析通道建立后的内膜增生.

Objective To study the effecof erythropoietin on vasculasmooth muscle cell proliferation and apoptosis.MethodMethyl thiazol tetrazolium (MTT) and 5-Ethynyl-2'-deoxyuridine (EdU) assaywere used to detecthe effecof erythropoietin on smooth muscle cell proliferation.TdT-mediated dUTP nick end labeling (TUNEL) staining waused to detecthe effecof erythropoietin on smooth muscle cell apoptosis.ResultMTresultshowed thaerythropoietin could inhibismooth muscle cell proliferation which decreased by 55％ (P ＜ 0.05).Extracellularegulated protein kinase(ERK) inhibitocould partly reverse the inhibition of erythropoietin on smooth muscle cell proliferationwhich increased by 42.4％ (P ＜ 0.05).EdU assay resultshowed thaerythropoietin could inhibismooth muscle cell proliferation which decreased by 49.6％, and ERK inhibitoincreased cell proliferation rate by 44.6％ (P ＜ 0.05).TUNEL staining resultshowed thaerythropoietin effectively promoted smooth muscle cell apoptosis, which wa3.14 timehighethan in control group (P ＜ 0.05).ERK inhibitohad no function in thiproces(P ＞ 0.05).Conclusion Ouresearch indicated thaerythropoietin mighinhibiintimal hyperplasiof hemodialysiaccesthrough inhibiting smooth muscle cell proliferation and promoting theiapoptosis.
