目的 观察经尾静脉、腹膜腔或肝内注射3种不同途径移植骨髓间充质干细胞(B MSCs)对大鼠肝纤维化进程的影响.方法 建立四氯化碳(CCl4)诱导的大鼠肝纤维化模型,第6周时BMSCs分别经尾静脉、腹膜腔或肝内注射途径回输.造模12周结束时评价大鼠存活率、腹水发生、炎症及纤维化.结果 造模12周结束时4组(n=16)生存率差异无统计学意义(x2=0.82,P >0.05),但腹水发生率差异有统计学意义(x2=9.019,P<0.05).仅经尾静脉移植组炎症活动度、纤维化以及α-平滑肌肌动蛋白(α-SMA)表达均低于对照组(P<0.01).结论 经外周静脉移植BMSCs能减轻大鼠肝脏炎症和α-SMA表达、减轻肝纤维化.

Objective To investigate the effecof the bone narrow mesenchymal stem cell(BMSCs) transplantation vidifferenadministration routein the treatmenof carbon tetrachloride (CCl4)-induced livefibrosiin rats.MethodThe hepatifibrosimodelwere induced by CCl4 in rats.The BMSCof the forth generation (5 × 1010/L) were injected into the livedirectly, viintraperitoneal injection ovithe caudalivenon the sixth week of the hepatifibrosimodeling in rats.Six weekafteBMSCtransplantation, the pathological changeof the livewere evaluated by Scheueinflammation activity score and Schmid M fibrosigrading score respectively.Also, the activation stage of HSin the livewadetected by immunohistochemistry and Western blotting.ResultSix weekafteBMSCtransplantation, there were obviously differencein asciteincidence among fougroup(x2 =9.019, P < 0.05).Acompared with the control group, the inflammation activity grade, fibrosigrade and α-smooth muscle actin (α-SMA) expression in the group with BMSCtranplantation vithe caudalivenwere significantly decreased (P < 0.01).Conclusion BMSCtransplantation vithe caudalivencould alleviate inflammation reaction,lessen the activation stage of HSin the liveand improve fibrosistage in transplanted ratwith livecirrhosiinduced by CCl4.
