目的 探讨丙戊酸钠逆转人肺癌细胞株A549/DDP顺铂耐药性的体外研究及其机制.方法 以浓度为10 mol/L的顺铂联合3 mmol/L的丙戊酸钠干预A549/DDP细胞株,采用噻唑蓝(MTT)法检测细胞增殖;膜联蛋白Ⅴ/碘化丙锭(Annexin Ⅴ/PI)双染法检测细胞早期凋亡;反转录聚合酶链反应(RT-PCR)检测细胞凋亡因子Survivin mRNA表达水平.结果 顺铂单药对A549/DDP的生长抑制作用较弱,而丙戊酸钠单药可有效抑制A549/DDP细胞的增殖,顺铂单药组干预A549/DDP细胞48 h的早期凋亡率为(7.84±1.34)%,而丙戊酸钠单药组为(23.29±1.98)%,两药联合组为(45.12±3.26)%.顺铂单药组Survivin mRNA水平表达无明显改变,而丙戊酸钠单药组及两药联合组的Survivin mRNA表达水平均明显下调,各项检测指标联合用药组与单药组比较差异均有统计学意义(P<0.05),且两药联合有协同作用,丙戊酸钠可逆转A549/DDP细胞株对顺铂的耐药性.结论 丙戊酸钠可逆转A549/DDP对顺铂的耐药性,其机制可能与下调Survivin的表达水平相关.

목적 탐토병무산납역전인폐암세포주A549/DDP순박내약성적체외연구급기궤제.방법 이농도위10 mol/L적순박연합3 mmol/L적병무산납간예A549/DDP세포주,채용새서람(MTT)법검측세포증식;막련단백Ⅴ/전화병정(Annexin Ⅴ/PI)쌍염법검측세포조기조망;반전록취합매련반응(RT-PCR)검측세포조망인자Survivin mRNA표체수평.결과 순박단약대A549/DDP적생장억제작용교약,이병무산납단약가유효억제A549/DDP세포적증식,순박단약조간예A549/DDP세포48 h적조기조망솔위(7.84±1.34)%,이병무산납단약조위(23.29±1.98)%,량약연합조위(45.12±3.26)%.순박단약조Survivin mRNA수평표체무명현개변,이병무산납단약조급량약연합조적Survivin mRNA표체수평균명현하조,각항검측지표연합용약조여단약조비교차이균유통계학의의(P<0.05),차량약연합유협동작용,병무산납가역전A549/DDP세포주대순박적내약성.결론 병무산납가역전A549/DDP대순박적내약성,기궤제가능여하조Survivin적표체수평상관.
Objective To investigate the effecand mechanism of valproate on reversing cisplatin-resistance of lung cancecell line A549/DDP.MethodCisplatin, valproate were tested alone and in combination fotheiabilitieto inhibiA549/DDP cell line proliferation by methyl thiazol tetrazolium (MTT).The early apoptosiof A549/DDP cell line were detected by Annexin Ⅴ/propidium iodide (PI) double staining method, and the expression of Survivin mRNwere detected by reverse transcriptase-polymerase chain reaction (RT-PCR).ResultCisplatin could inhibited faintly the proliferation of A549/DDP cell line, however, valproate and combination could inhibited effectively the proliferation of A549/DDP cell line.The early stage apoptosirate of A549/DDP cellwa(7.84 ± 1.34) % when treated with cisplatin, and wa(23.29 ±1.98)% when treated with valproate, and wa(45.12 ±3.26)% when they were treated with cisplatin combined with valproate.The expression of Survivin mRNhad no significanchangein cisplatin group.The expression of Survivin mRNlevelwere down-regulated in valproate and combination group.Comparisonof treated groupand control group have significandifference (P < 0.05)and comparisonof combination groupand single drug group have also significandifference (P < 0.05).Conclusion Valproate could reverse the drug resistance to cisplatin in A549/DDP cell line.The potential mechanism idown-regulated Survivin mRNlevelto promote apoptosiof cells.
