目的:探讨不同前处理方法、蒽酮硫酸显色法和苯酚硫酸显色法对粗多糖分析结果的影响。方法:以粗多糖的含量为评价指标,对灵芝孢子粉、乌药精粉应用不同的提取方法和测定方法进行分析。结果:不同的前处理方法测定灵芝孢子粉、乌药精粉中的粗多糖的含量不同;相同前处理下,采用苯酚硫酸显色法和蒽酮硫酸显色法,样本中粗多糖的含量无差异。结论:不同样本需采用不同的前处理,否则会出现样本粗多糖含量过高或过低现象;蒽酮硫酸显色法和苯酚硫酸显色法所测粗多糖线性、稳定性较好,适合大多数样品的检测。

OBJECTIVE:To probe into the effect of different pre-treatment methods and different determination methods like anthrone-sulfuric acid method and phenol-sulfuric acid method on the analysis of crude polysaccharide. METHODS:The content of crude polysaccharide was set as the evaluation index, different extraction methods and determination methods of ganoderma lucidum spores powder and lindera aggregata flavor powder were studied. RESULTS: Different contents of crude polysaccharide were determined in ganoderma lucidum spores powder and lindera aggregata flavor powder by different pre-treatment methods; to adopt the phenol-sulfuric acid method and phenol-sulfuric acid method with the same pre-treatment methods, there were no difference of the contents of crude polysaccharide in the samples.CONCLUSIONS: Different samples need to adopt different pre-treatment methods, otherwise the determined contents of crude polysaccharide would be too high or too low in the samples; phenol -sulfuric acid method and anthrone-sulfuric acid anthrone method in determination of crude polysaccharide has good linear correlation and stability, which is suitable for most detection of the samples.
