为进一步提高终极腐霉EPA产量,在初步优化发酵条件的基础上,通过Plackett-Burman和Box-Behnken实验设计优化终极腐霉生产 EPA 发酵工艺. 获得的最优发酵工艺条件为:蔗糖8%,硝酸钾0. 4%,酵母粉0. 65%,磷酸氢二钠0. 175%,硫酸镁0. 065%,大豆油1. 0%,起始pH 6. 0,装液量50 mL/250 mL. 在最优发酵工艺条件下,EPA产量达到541. 61 mg/L,比初步优化产量(456. 39 mg/L)提高了85. 22 mg/L.

In order to increase the EPA yield produced by Pythium ultimum, the fermentation process of Pythium ultimum for EPA production was optimized by Plackett-Burman design combined with Box-Behnken design on the basis of preliminary optimal fermentation conditions. The results showed that the optimal fermentation conditions were obtained as follows: sucrose 8%, KNO3 0. 4%, yeast powder 0. 65%, Na2 HPO4 0. 175%, MgSO4 0. 065%, soybean oil 1. 0%, initial pH 6. 0, liquid volume in flask 50 mL/250 mL. Under the optimal conditions, EPA yield reached 541. 61 mg/L, which increased by 85. 22 mg/L compared with that of preliminary optimization (456. 39 mg/L) .
