目的:研究福林法测定蛋白酶酶活时的影响因素。方法测定2709碱性蛋白酶酶活,对比分析了各种常见差异造成的影响。结果酪蛋白、滤纸的选择及样品稀释液配制时间对酶活测定影响较大,福林试剂、样品溶解方式等影响不明显,而酶活计算方法的不同也会带来差异。结论检测人员在检测前应明确采用的方法、试剂及计算方法。

ObjectiveTo study the influencing factors of activity determination for protease by Folin method.Methods The alkaline protease fromBacillus licheniformis (NO.2709) was taken as sample. Several frequent differences involved in practical operation were investigated in the research.Results The determination was influenced greatly by the casein, filter paper and blending time of sample diluent, meanwhile insignificantly by Folin-Ciocalteu’s phenol reagent and dissolving process of samples. Alternative calculations were considered to lead to different results.Conclusion The operation details, involving determination method, reagents and calculation method, must be confirmed before determination.
