目的:采用SAX-HPLC快速鉴别不同种属来源的粗品肝素钠。方法通过肝素酶酶解不同种属来源的粗品肝素钠,以其酶解后的二糖组份为考察对象,通过SAX HPLC法比较二糖的相对组成,鉴定种属来源。同时在猪来源肝素中分别添加不同比例(5%,10%,15%,20%,25%)的牛来源肝素或羊来源肝素,经肝素酶酶解后,通过SAX HPLC法鉴别在猪来源肝素中是否能快速检出不同种属来源的其他肝素。结果对于不同种属来源的粗品肝素钠,它们的二糖组成具有显著差异;若猪来源肝素混入5%羊来源肝素,即可通过其二糖组成判断。结论此法操作简便,成本较低,可用于不同种属来源的粗品肝素钠的定性检测。

Objective To identify the origin of species of crude heparin sodium by SAX-HPLC.Methods With the digested disaccharide components as the study object which was got through heparinase hydrolysis of crude heparin sodium, SAX-HPLC was used to identify the origin of species by the comparison of disaccharide relative composition. Meanwhile, bovine-or ovine-sourced heparin was added to the porcine crude heparin at different proportions (5 %,10 %,15 %,20 %,25 %). After the digestion by heparinase, it was confirmed whether heparin from foreign species could be detected quickly within porcine crude heparin by SAX-HPLC.ResultsThere were significant differences on disaccharide composition among crude heparin from different species. If porcine crude heparin was mixed with 5 % ovine crude heparin, it could be detected by the disaccharide composition.Conclusion This method is simple and low-cost, and can be used for the qualitative determination of crude heparin sodium from different species.
