目的:采用高效液相色谱-蒸发光散射法分析确定人体以及七种动物油脂的甘油三酯种属特征。方法用丙酮溶解19种甘油三酯标准品,分析这19种标准甘油三酯特征峰,色谱柱条件为 C18色谱柱,流动相是二氯甲烷-乙腈-冰乙酸(30:70:0.2,V/V/V),流速1.0 mL/min,采用蒸发光散射检测器检测,以此得出特征标准峰,再对样品进行分析;采集人体、七种动物(猪、牛、羊、鸡、鸭、鹅、鱼)油脂,丙酮溶解后直接采用与标准品相同的条件进样分析,分离各种甘油三酯,对比混合标准样品特征峰处理各种谱图数据。结果人体油脂、七种动物油脂的谱图数据特征性强,能够区分人以及这七种动物。结论甘油三酯可以作为鉴定人类和这七种动物油脂种属的依据,为案件侦破指明方向。

Objective To explore the species characteristics of fat from human and seven kinds of animals (pigs, cattle, sheep, chickens, ducks, geese, fish) by the analysis of triglycerides using high performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD). Methods 19 kinds of triglycerides standards were dissolved in acetone and analyzed by HPLC-ELSD. Chromatography was achieved using a C18 column and an isocratic mobile phase consisting of methylene chloride/acetonitrile/ice acetic acid (30/70/0.2, V/V/V) at a flow rate of 1 mL/min, and ELSD detection was performed at 70°C for the drift tube. Fat of human and seven kinds of animals was subjected to the same treatment as that of triglycerides standards in identical conditions, and the resultant chromatograms being compared with those of triglycerides standards to determine the types of triglyceride. Results Human fat has 9 types of triglyceride, and the fat from pig, cattle, sheep, chicken, duck, goose and fish has 10, 14, 13, 12, 11, 14, 15 types of triglycerides, respectively. All of these fats have their own special triglyceride types as their species’ characteristics. For the fats of same species animals raised in different area, the types of triglyceride, though various in content, have shown the same characteristics. Conclusions Types of triglycerides are reliable for identifying whether the fat is from human and seven kinds of animals tested in this study, and this can provide useful information for case investigation.
