低温保存造血干细胞的效果受诸多因素的影响,如温度、降温速率、细胞含量、低温保护剂、解冻复温过程、保存时间以及检测指标,由于各研究中心之间所用方法存在一定差异,导致结果可比性差,循证力度较弱。因此至今为止,国内尚无相关操作指南,国外也只有欧洲的国际细胞治疗学会和血液及骨髓移植组联合认证委员会指南。作者结合自身多年血细胞低温保存工作经验和各种文献,起草了适用于医院小型低温保存库的操作规程,供同行参考,并期待大家批评指正。

The efficacy of cryopreservation of hematopoietic stem cells was influenced by a lot of factors, such as temperature, cooling rate, cell concentration, cryopreservative, thawing proce-dure, preserved period and measurement of cell functionalities.There were some differences in tech-nology and methodology between research centers in the world, so the results from their publications could be poorly compared with each other and had a weak potency in evidence-based medicine.Ex-cept for the established protocol for cryostorage of mobilized peripheral blood ( MPB) hematopoietic stem cells/hematopoietic progenitor cells ( HSC/HPC) and bone marrow ( BM) HSC/HPC in Joint Accreditation Committee of International Stem Cell Transplantation ( ISCT ) Europe and European Bone Marrow Transplantation ( EBMT) ( JACIE) guidelines, there was no guideline in China and the world.With the help of our experiences ( for many years in cryomedical field) and related litera-tures we have drawn up this protocol for cryopreservation and thawing of hematopoietic stem cells suited for petty bank in hospital.Any comments or revisions will be welcome.
