目的:探讨人胎儿卵泡的发育形成过程中卵细胞凋亡及其调控机制。方法收集2013年1~12月广东省计划生育科学技术研究所妊娠24~32周引产的女性胎儿卵巢组织18例,其中24~27+6周胎儿卵巢8例,28~32周胎儿卵巢10例。采用免疫组织化学染色法观察胎儿卵巢卵泡发育及形成过程中凋亡因子Caspase-3、TNF-α、bcl-2、bax的表达情况。结果 Caspase-3表达在孕中期及孕晚期表达均为强阳性,TNF-α在卵母细胞中均表达,胎儿卵巢内的颗粒细胞及其他间质组织细胞未见阳性表达。凋亡抑制因子bcl-2的表达在孕中期及孕晚期均为阴性。促凋亡因子Bax在卵母细胞及卵巢颗粒细胞内均为阳性表达。结论胎儿卵巢发育过程中,原始卵母细胞不断发生凋亡,由四种凋亡因子同时共同参与,而bcl-2和bax可能通过活化Caspase-3引起少量颗粒细胞凋亡。

목적:탐토인태인란포적발육형성과정중란세포조망급기조공궤제。방법수집2013년1~12월광동성계화생육과학기술연구소임신24~32주인산적녀성태인란소조직18례,기중24~27+6주태인란소8례,28~32주태인란소10례。채용면역조직화학염색법관찰태인란소란포발육급형성과정중조망인자Caspase-3、TNF-α、bcl-2、bax적표체정황。결과 Caspase-3표체재잉중기급잉만기표체균위강양성,TNF-α재란모세포중균표체,태인란소내적과립세포급기타간질조직세포미견양성표체。조망억제인자bcl-2적표체재잉중기급잉만기균위음성。촉조망인자Bax재란모세포급란소과립세포내균위양성표체。결론태인란소발육과정중,원시란모세포불단발생조망,유사충조망인자동시공동삼여,이bcl-2화bax가능통과활화Caspase-3인기소량과립세포조망。
Objective To investigate the apoptosis and the expression of apoptosis related protein Caspase-3, TNF-α, bcl-2, bax of the oocyte during human fetal development. Methods Ovarian tissues from 18 fetuses (aged from 24 to 32 weeks) were obtained after labortion induced by mifepristone and PG from January to December 2013 in Guangdong Family Planning Research Institute, including 8 fetuses of midpregnancy (aged 24-27+6 weeks) and 10 fetuses of late-pregnancy (aged 28-32 weeks). The morphology and development of oocyte were observed using HE-staining and im-munohistochemistry. The expression of the apoptosis related protein, such as Caspase-3, TNF-α, bcl-2, bax of the oocyte during human fetal follicular development were examined. Results The Caspase-3 was positive both in the fetal oocytes and the granulosa cells. TNF-α was positive in the oocytes. Bcl-2 was negative both in the fetal oocytes and the granulosa cells. Bax was positive both in the fetal oocytes and the granulosa cells. Conclusion The four apoptosis factors are all participate in the oocyte apoptosis during human fetal development. bcl-2 and bax may result in the granulosa cells apoptosis participate through activate the Caspase-3.
