目的:观察三黄糖肾康颗粒对2型糖尿病大鼠骨组织肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6表达的影响。方法取雌性Wistar大鼠,正常对照组10只,制作骨质疏松模型10只和糖尿病模型50只。糖尿病模型大鼠进一步分成模型对照组,中药预防、高、低剂量组和钙尔奇碳酸钙D3(西药)组,每组10只。7组分别给予相应干预,喂养20周后取材,全自动生化仪检测空腹血糖(FBG);酶联免疫吸附法(ELISA)检测血清TNF-α和IL-6水平;双能X线骨密度仪(DXA)检测骨密度,实时定量聚合酶链反应(RT-PCR)检测骨组织TNF-α和IL-6 mRNA水平;蛋白免疫印迹法(Western blot)检测骨组织TNF-α和IL-6蛋白水平。结果与骨质疏松模型组比较,中药预防、高、低剂量组及西药组骨密度升高,血清TNF-α、IL-6下降,骨组织IL-6 mRNA及蛋白表达下降(P<0.05),中药预防、高剂量组和西药组骨组织TNF-αmRNA及蛋白表达下降(P<0.05)。与糖尿病模型对照组比较,中药预防、高、低剂量组及西药组FBG、血清TNF-α、IL-6均明显下降(P<0.05),中药预防组、中药高剂量组和西药组骨组织TNF-αmRNA及蛋白表达下降(P<0.05),中药预防、低剂量和西药组骨组织IL-6 mRNA及蛋白表达下降(P<0.05)。结论糖尿病大鼠血清和骨组织TNF-α、IL-6表达增高,三黄糖肾康颗粒能够改善糖尿病大鼠的炎症状况。

목적:관찰삼황당신강과립대2형당뇨병대서골조직종류배사인자(TNF)-α、백세포개소(IL)-6표체적영향。방법취자성Wistar대서,정상대조조10지,제작골질소송모형10지화당뇨병모형50지。당뇨병모형대서진일보분성모형대조조,중약예방、고、저제량조화개이기탄산개D3(서약)조,매조10지。7조분별급여상응간예,위양20주후취재,전자동생화의검측공복혈당(FBG);매련면역흡부법(ELISA)검측혈청TNF-α화IL-6수평;쌍능X선골밀도의(DXA)검측골밀도,실시정량취합매련반응(RT-PCR)검측골조직TNF-α화IL-6 mRNA수평;단백면역인적법(Western blot)검측골조직TNF-α화IL-6단백수평。결과여골질소송모형조비교,중약예방、고、저제량조급서약조골밀도승고,혈청TNF-α、IL-6하강,골조직IL-6 mRNA급단백표체하강(P<0.05),중약예방、고제량조화서약조골조직TNF-αmRNA급단백표체하강(P<0.05)。여당뇨병모형대조조비교,중약예방、고、저제량조급서약조FBG、혈청TNF-α、IL-6균명현하강(P<0.05),중약예방조、중약고제량조화서약조골조직TNF-αmRNA급단백표체하강(P<0.05),중약예방、저제량화서약조골조직IL-6 mRNA급단백표체하강(P<0.05)。결론당뇨병대서혈청화골조직TNF-α、IL-6표체증고,삼황당신강과립능구개선당뇨병대서적염증상황。
Objective To observe the effects of Sanhuangtangshenkang particles on the expression levels of tumor ne?crosis factor (TNF)-α, interleukin (IL)-6 in bone tissue of type 2 diabetic (T2DM) rat model. Methods There were seven groups of rats in this study. Ten female Wistar rats were used as control. Ten rats were established for osteoporosis model, and fifty rats were established for diabetic model. Fifty diabetic model rats were further divided into model control group, tra?ditional Chinese medicine (TCM) prevention group (high-dose TCM group, low-dose TCM group) and Western medicine group, with 10 rats for each group. All rats were given the corresponding intervention for 20 weeks. Automatic biochemistry analyzer was used to determine fasting blood glucose (FBG). ELISA was used to detect serum TNF-αand IL-6. Dual-energy X-ray absorptiometry (DXA) was used to assess bone density. Real-time PCR was used to appraise the mRNA of TNF-αand IL-6 in bone tissue. Western blot assay was used to measure protein expressions of TNF-αand IL-6 in bone tissue. Re?sults Compared with osteoporosis model group, bone density was significantly increased, serum levels of TNF-αand IL-6, the mRNA and protein expression level of IL-6 were significantly decreased in bone tissue of TCM groups and Western medi?cine group (P<0.05). The expression levels of TNF-αmRNA and protein were significantly decreased in bone tissue of in high-dose TCM prevention group and Western medicine group (P<0.05). Compared with the diabetic model group, the se?rum levels of FBG, TNF-αand IL-6 were significantly decreased in TCM groups and Western medicine group (P<0.05). The expression levels of TNF-αmRNA and protein were significantly decreased in high-dose TCM prevention group and Western medicine group (P<0.05). The mRNA and protein expression levels of IL-6 were significantly decreased in low-dose TCM prevention group and Western medicine group (P<0.05). Conclusion The expression levels of TNF-αand IL-6 are increased in serum and bone tissue of diabetic rats. Sanhuangtangshenkang particles can improve the inflammatorystate to play prevention and therapeutic effect on diabetic rats.
