目的 探讨GABA A受体( GABAa受体)激动剂对新生期大鼠海马神经元癫痫样放电的影响.方法 出生24 h内新生SD大鼠,取海马神经元原代培养至第9 d,用"无镁细胞外液"孵育3 h,建立体外癫痫样放电模型,使用全细胞膜片钳技术记录GABAa受体激动剂(氯硝西泮、蝇蕈醇、异孕烷醇酮)对新生大鼠海马神经元癫痫放电的作用. 结果 通过膜片钳的记录,体外培养的新生大鼠海马神经元在"无镁细胞外液"的诱导下可形成稳定的癫痫样放电. 氯硝西泮干预后神经元放电的幅度和频率与干预前比较差异无统计学意义( P>0.05). 蝇蕈醇和异孕烷醇酮干预后神经元放电的幅度和频率与干预前比较明显降低(均P<0.05). 结论 GABAa受体激动剂在新生大鼠海马神经元体外癫痫样放电模型中介导抑制作用,其中蝇蕈醇和异孕烷醇酮可以快速、明显抑制新生大鼠癫痫样放电.

목적 탐토GABA A수체( GABAa수체)격동제대신생기대서해마신경원전간양방전적영향.방법 출생24 h내신생SD대서,취해마신경원원대배양지제9 d,용"무미세포외액"부육3 h,건입체외전간양방전모형,사용전세포막편겸기술기록GABAa수체격동제(록초서반、승심순、이잉완순동)대신생대서해마신경원전간방전적작용. 결과 통과막편겸적기록,체외배양적신생대서해마신경원재"무미세포외액"적유도하가형성은정적전간양방전. 록초서반간예후신경원방전적폭도화빈솔여간예전비교차이무통계학의의( P>0.05). 승심순화이잉완순동간예후신경원방전적폭도화빈솔여간예전비교명현강저(균P<0.05). 결론 GABAa수체격동제재신생대서해마신경원체외전간양방전모형중개도억제작용,기중승심순화이잉완순동가이쾌속、명현억제신생대서전간양방전.
Objective To observe the effects of GABA receptor A ( GABAa) agonists on hippocampal neurons epileptiform discharge in neonate rat.Methods Neonatal SD rats (<24 h old) were adopted to draw hippocampal neurons for primary culture.On day 9, seizure like events were induced by lowering magnesium concentration.Then, the discharge activity were recorded by whole-cell patch clamp.The effects of GABAa agonists ( Clonazepam, Muscimol and allopregnanolone) were analyzed.Results By the recording of whole-cell patch clamp, there were stabile epileptic discharges of hippocampal neurons of neonate rats exposed to magnesium-free extracelhlar fluid. Compared with before the intervention, the amplitude and frequency of the discharge of neurons after the intervention of Clonazepam were no significantly difference (P>0.05).Compared with before the intervention, the amplitude and frequency of the discharge of neurons after the intervention of Muscimol and allopregnanolone were significantly decreased (all P<0.05).Conclusions GABAa agonists is mediated inhibition effect in epileptic discharge modle of hippocampal neurons of neonate rats.Muscimol and allopregnanolone can rapidily and obviously inhibit the epilepsy discharges.
