目的:探讨大鼠小肠对胶原蛋白口服微球的吸收作用。方法取250~350 g 5个月龄 Wistar 大鼠,建立小肠外翻肠囊模型和单向肠灌流模型,采用二喹啉甲酸(BCA)蛋白定量分析试剂盒检测胶原蛋白在小肠各段的吸收情况。结果不同粒径海藻酸钙/壳聚糖(SA/ CTS)微球在外翻肠囊模型各小肠道累积吸收:500μm 微球<200μm 微球<10μm 微球(P<0.01,除空肠外)。10μm 微球累积透过吸收:SA/ CTS 微球>SA 微球(十二指肠,空肠 P<0.05;回肠 P<0.01);单向肠灌流模型 SA/ CTS 微球吸收速率常数(Ka )和有效透过系数(kef )均显著性高于 SA 微球(空肠段 P<0.01)。结论胶原蛋白微球的小肠吸收与粒径有相关性,SA/ CTS 微球中生物黏附材料壳聚糖有助于胶原蛋白在小肠各段的吸收。
目的:探討大鼠小腸對膠原蛋白口服微毬的吸收作用。方法取250~350 g 5箇月齡 Wistar 大鼠,建立小腸外翻腸囊模型和單嚮腸灌流模型,採用二喹啉甲痠(BCA)蛋白定量分析試劑盒檢測膠原蛋白在小腸各段的吸收情況。結果不同粒徑海藻痠鈣/殼聚糖(SA/ CTS)微毬在外翻腸囊模型各小腸道纍積吸收:500μm 微毬<200μm 微毬<10μm 微毬(P<0.01,除空腸外)。10μm 微毬纍積透過吸收:SA/ CTS 微毬>SA 微毬(十二指腸,空腸 P<0.05;迴腸 P<0.01);單嚮腸灌流模型 SA/ CTS 微毬吸收速率常數(Ka )和有效透過繫數(kef )均顯著性高于 SA 微毬(空腸段 P<0.01)。結論膠原蛋白微毬的小腸吸收與粒徑有相關性,SA/ CTS 微毬中生物黏附材料殼聚糖有助于膠原蛋白在小腸各段的吸收。
목적:탐토대서소장대효원단백구복미구적흡수작용。방법취250~350 g 5개월령 Wistar 대서,건립소장외번장낭모형화단향장관류모형,채용이규람갑산(BCA)단백정량분석시제합검측효원단백재소장각단적흡수정황。결과불동립경해조산개/각취당(SA/ CTS)미구재외번장낭모형각소장도루적흡수:500μm 미구<200μm 미구<10μm 미구(P<0.01,제공장외)。10μm 미구루적투과흡수:SA/ CTS 미구>SA 미구(십이지장,공장 P<0.05;회장 P<0.01);단향장관류모형 SA/ CTS 미구흡수속솔상수(Ka )화유효투과계수(kef )균현저성고우 SA 미구(공장단 P<0.01)。결론효원단백미구적소장흡수여립경유상관성,SA/ CTS 미구중생물점부재료각취당유조우효원단백재소장각단적흡수。
Objective To investigate small intestinal absorption behavior of oral collagen peptides microspheres in rats. Methods The everted gut sac (EGS) and Single-pass intestinal perfusion (SPIP) model were established in Wistar rats (250-350 g, 5 months).The absorption of collagen peptide in different segments of rat small intestines was detected by BCAmethod. Results The cumulative transmittance of different particle size of calcium alginate/ Chitosan (SA/ CTS) microspheresin EGS experiments was that 500 μm microspheres<200 μm microspheres<10 μm microspheres ( P<0.01, except the jejunumintestine).The cumulative transmittance of microspheres ( 10 μm) was: SA/ CTS rate ( Ka ) and permeability rate ( Peff )microspheres> SA microspheres(duodenum segment, jejunum segment P<0.05;ileum segment, P<0.01).The absorption rate ofSA/ CTS microspheres was evidently higher than that of SA microspheres in SPIP experiments. Conclusion Small intestineabsorption of collagen peptides microspheres is correlated with particle size, and the chitosan in SA/ CTS microspheres isbeneficial to the absorption of collagen peptide in small intestine segments.
