目的 探讨慢性阻塞性肺疾病(COPD)急性加重期患者外周血中干扰素γ诱导蛋白10(IP-10)的表达及意义.方法 收集2012年7月至2013年10月广州医科大学附属第三医院治疗的COPD急性加重期患者58例作为观察组,另纳入同期28名健康体检者作为对照组.用TRIzol试剂提取细胞总mRNA,用反转录试剂盒将RNA反转录为cDNA.用实时荧光定量聚合酶链反应检测IP-10 mRNA水平,酶联免疫吸附试验检测IP-10蛋白质表达水平.Pearson相关检验分析IP-10表达水平与肺功能的相关性.结果 COPD急性加重期患者外周血中IP-10 mRNA及蛋白质表达水平均明显高于对照组[(12.3±4.3)比(3.8 ±1.9),(177±35) ng/L比(87±31) ng/L],差异有统计学意义(P<0.05);IP-10表达水平与第1秒用力呼气容积、用力肺活量、第1秒用力呼气容积/用力肺活量呈负相关(r=-0.543、-0.486、-0.471,均P<0.05).结论 COPD急性加重期患者外周血中IP-10表达水平增高,且IP-10表达水平与肺功能呈负相关,可能参与COPD的发病.

목적 탐토만성조새성폐질병(COPD)급성가중기환자외주혈중간우소γ유도단백10(IP-10)적표체급의의.방법 수집2012년7월지2013년10월엄주의과대학부속제삼의원치료적COPD급성가중기환자58례작위관찰조,령납입동기28명건강체검자작위대조조.용TRIzol시제제취세포총mRNA,용반전록시제합장RNA반전록위cDNA.용실시형광정량취합매련반응검측IP-10 mRNA수평,매련면역흡부시험검측IP-10단백질표체수평.Pearson상관검험분석IP-10표체수평여폐공능적상관성.결과 COPD급성가중기환자외주혈중IP-10 mRNA급단백질표체수평균명현고우대조조[(12.3±4.3)비(3.8 ±1.9),(177±35) ng/L비(87±31) ng/L],차이유통계학의의(P<0.05);IP-10표체수평여제1초용력호기용적、용력폐활량、제1초용력호기용적/용력폐활량정부상관(r=-0.543、-0.486、-0.471,균P<0.05).결론 COPD급성가중기환자외주혈중IP-10표체수평증고,차IP-10표체수평여폐공능정부상관,가능삼여COPD적발병.
Objective To explore the expression and significance of interferon-γ induced protein 10 (IP-10) in serum of patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD).Methods Fifty-eight patients with AECOPD (observation group) and 28 healthy subjects (control group) from July 2012 to October 2013 were enrolled.The total mRNA in peripheral blood was extracted by TRIzol.Then RNA was reversely transcribed into cDNA by using reverse transcription Kit, subsequently, the mRNA level of IP-10 was detected by real-time polymerase chain reaction (PCR).The expression level of IP-10 protein in serum was detected by enzyme linked immunosorbent assay method.The relation between IP-10 and pulmonary function was analyzed by Pearson's correlation analysis.Results The RNA and protein level of IP-10 in observation group were significantly higher than those in control group [(12.3 ± 4.3) vs (3.8 ± 1.9), (177 ± 35) ng/L vs (87 ± 31) ng/L] (P <0.05).The IP-10 level in serum was negatively correlated with forced expiratory volume in one second (FEV1), forced vital capacity (FVC) and ratio of FEV1 and FVC in percentage (FEV1/FVC) (r =-0.543,-0.486,-0.471, P < 0.05).Conclusion The expression level of IP-10 in serum is increased in patients with AECOPD;the IP-10 level in serum is negatively correlated with pulmonary function and can be involved in the pathogenesis of AECOPD.
