目的研究血浆高分子量激肽原(high molecular weight kininogen ,HK)在大肠杆菌导致的脓毒症中的作用。方法生长到对数期的大肠杆菌用PBS清洗后重悬,对高分子量激肽原的基因敲除小鼠Kng1-/-小鼠和野生型Kng1+/+小鼠分别腹腔注射大肠杆菌,构建细菌感染模型,并观察小鼠的生存率;应用血平板对血液中细菌计数;H&E染色观察肺脏的病理变化;应用ELISA检测血浆中炎症因子蛋白水平的表达;应用实时定量PCR检测白细胞中炎症因子mRNA水平的表达。结果在大肠杆菌感染导致的脓毒症模型中,Kng1基因的敲除促进了小鼠死亡;Kng1-/-小鼠血液中细菌数目明显高于Kng1+/+小鼠;Kng1-/-小鼠肺部病理切片与Kng1+/+小鼠相比损伤严重;Kng1-/-小鼠血浆中炎症因子蛋白水平的表达与白细胞中炎症因子mRNA水平的表达均高于Kng1+/+小鼠。结论血浆高分子量激肽原在大肠杆菌导致的感染中起保护作用。

목적연구혈장고분자량격태원(high molecular weight kininogen ,HK)재대장간균도치적농독증중적작용。방법생장도대수기적대장간균용PBS청세후중현,대고분자량격태원적기인고제소서Kng1-/-소서화야생형Kng1+/+소서분별복강주사대장간균,구건세균감염모형,병관찰소서적생존솔;응용혈평판대혈액중세균계수;H&E염색관찰폐장적병리변화;응용ELISA검측혈장중염증인자단백수평적표체;응용실시정량PCR검측백세포중염증인자mRNA수평적표체。결과재대장간균감염도치적농독증모형중,Kng1기인적고제촉진료소서사망;Kng1-/-소서혈액중세균수목명현고우Kng1+/+소서;Kng1-/-소서폐부병리절편여Kng1+/+소서상비손상엄중;Kng1-/-소서혈장중염증인자단백수평적표체여백세포중염증인자mRNA수평적표체균고우Kng1+/+소서。결론혈장고분자량격태원재대장간균도치적감염중기보호작용。
Objective To determine the role of plasma high molecular weight kininogen ( HK) in bacte-rial infection .Methods Mice received peritoneal injection with E .Coli to induce infection .The survival rate was observed ,The bacteria colony forming unit ( CFU ) of blood was counted .The H&E staining of lung , the protein level of inflammatory cytokines in plasma and their mRNA level in white blood cell were compared be -tween Kng1 +/+mice and Kng1 -/-mice.Results After injected with E.Coli,Kng1 deficiency increased mor-tality,Kng1 -/-mice had a decrease in bacteria clearance in blood and deteriorated lung injury compared with Kng1 +/+mice;Furthermore ,the deficiency of kininogen increased plasma cytokines protein levels and mRNA levels in white blood cells .Conclusion Plasma high molecular weight kininogen has a protective role in host defense against E .coli infection.
