目的 探讨吗啡预处理对离体心肌的保护作用. 方法 20只家猪随机分为空白组和吗啡组,每组10只,建立猪不停跳心脏离体模型,在心脏离体前30 min,吗啡组给予静脉注射吗啡0.3 mg/kg预处理,空白组给予同等剂量生理盐水,分别于心脏离体即刻、保存2h、 4h6、 h、8h时取右心室肌小梁制备组织匀浆. 采用羟安法检测心肌组织中超氧化物歧化酶( SOD)活性及丙二醇( MDA)、三磷酸腺苷( ATP)含量. 结果 两组动物均成功建立不停跳心脏离体模型. 随着实验时间延长 ,空白组和吗啡组心肌MDA含量均上升,而心肌SOD活性、ATP含量下降(P<0.05);在各时间点,吗啡组SOD活性、ATP含量高于空白组(P<0.05),MDA含量低于空白组(P<0.05). 透射电镜下观察发现两组心肌细胞有不同程度的细胞水肿、线粒体肿胀,但空白组出现较明显的心肌细胞、线粒体结构破坏,吗啡组心肌细胞、线粒体结构比较完整.结论 吗啡预处理对离体心肌有保护作用.
目的 探討嗎啡預處理對離體心肌的保護作用. 方法 20隻傢豬隨機分為空白組和嗎啡組,每組10隻,建立豬不停跳心髒離體模型,在心髒離體前30 min,嗎啡組給予靜脈註射嗎啡0.3 mg/kg預處理,空白組給予同等劑量生理鹽水,分彆于心髒離體即刻、保存2h、 4h6、 h、8h時取右心室肌小樑製備組織勻漿. 採用羥安法檢測心肌組織中超氧化物歧化酶( SOD)活性及丙二醇( MDA)、三燐痠腺苷( ATP)含量. 結果 兩組動物均成功建立不停跳心髒離體模型. 隨著實驗時間延長 ,空白組和嗎啡組心肌MDA含量均上升,而心肌SOD活性、ATP含量下降(P<0.05);在各時間點,嗎啡組SOD活性、ATP含量高于空白組(P<0.05),MDA含量低于空白組(P<0.05). 透射電鏡下觀察髮現兩組心肌細胞有不同程度的細胞水腫、線粒體腫脹,但空白組齣現較明顯的心肌細胞、線粒體結構破壞,嗎啡組心肌細胞、線粒體結構比較完整.結論 嗎啡預處理對離體心肌有保護作用.
목적 탐토마배예처리대리체심기적보호작용. 방법 20지가저수궤분위공백조화마배조,매조10지,건립저불정도심장리체모형,재심장리체전30 min,마배조급여정맥주사마배0.3 mg/kg예처리,공백조급여동등제량생리염수,분별우심장리체즉각、보존2h、 4h6、 h、8h시취우심실기소량제비조직균장. 채용간안법검측심기조직중초양화물기화매( SOD)활성급병이순( MDA)、삼린산선감( ATP)함량. 결과 량조동물균성공건립불정도심장리체모형. 수착실험시간연장 ,공백조화마배조심기MDA함량균상승,이심기SOD활성、ATP함량하강(P<0.05);재각시간점,마배조SOD활성、ATP함량고우공백조(P<0.05),MDA함량저우공백조(P<0.05). 투사전경하관찰발현량조심기세포유불동정도적세포수종、선립체종창,단공백조출현교명현적심기세포、선립체결구파배,마배조심기세포、선립체결구비교완정.결론 마배예처리대리체심기유보호작용.
Objective To investigate the protective effects of morphine preconditioning on myocardium in vitro.Methods Twenty pigs were randomly divided into blank group(n=10) and morphine group(n=10).A porcine beating-heart model in vitro was established.Thirty minutes before the porcine hearts were isolated,morphine group was given intravenous injection of morphine(0.3 mg/kg) for preconditioning,and blank group was given intravenously equal dose of normal saline.The ventricular trabeculae were taken out for tissue homogenate at the moment of heart isolation,2,4,6 and 8 hours after heart isolation,respectively.Chemical colorimetric assay was used to detect the activity of superoxide dismutase(SOD) of myocardium as well as the contents of malondialdehyde(MDA) and adenosine triphosphate(ATP) of myocardium.Results The porcine beating-heart model in vitro was established successfully in the two groups.In both of blank group and morphine group,the content of myocardial MDA increased,but the activity of myocardial SOD and the content of myocardial ATP decreased with the increasing experimental time(P<0.05).At all time points,morphine group showed higher activity of SOD,higher content of ATP and lower content of MDA compared with blank group(P<0.05).Cell edema and mitochondria swelling with different degrees were observed in myocardial cells under transmission electron microscope in the two groups.The structures of myocardial cells and mitochondria were damaged more obviously in the blank group,but the structures of myocardial cells and mitochondria were relatively complete in the morphine group.Conclusion Morphine preconditioning has protective effects on the myocardium in vitro.
