目的: Munc-18在中枢神经系统递质释放过程中具有重要作用,其控制着突触囊泡释放步骤的每一个环节。Munc-18功能异常与癫痫发病相关。本文主要探讨癫痫发作是否对小鼠海马内Munc-18蛋白的分布产生影响。方法将18只雄性成年昆明鼠分为三组,生理盐水组和癫痫组分别向腹腔注射生理盐水和海人藻酸(10mg/kg)溶液,对照组不做处理,观察小鼠的行为变化。观察结束后小鼠全部处死取脑,冰冻切片行免疫组织化学染色后观察Munc-18蛋白和c-Fos蛋白的表达情况。结果注射海人藻酸30min后小鼠癫痫样发作,首先出现胡须和嘴部抽动,接着翘尾、甩尾、点头、单侧上肢抽搐,达到4级标准。与对照组和生理盐水组比较,癫痫组小鼠海马区域的Munc-18蛋白分布减少同时伴有c-Fos蛋白的表达增加,以CA3和齿状回区域更加明显,差别具有统计学差异。结论癫痫能够导致Munc-18蛋白在海马区域分布减少,同时使该区域的神经兴奋性增高。
目的: Munc-18在中樞神經繫統遞質釋放過程中具有重要作用,其控製著突觸囊泡釋放步驟的每一箇環節。Munc-18功能異常與癲癇髮病相關。本文主要探討癲癇髮作是否對小鼠海馬內Munc-18蛋白的分佈產生影響。方法將18隻雄性成年昆明鼠分為三組,生理鹽水組和癲癇組分彆嚮腹腔註射生理鹽水和海人藻痠(10mg/kg)溶液,對照組不做處理,觀察小鼠的行為變化。觀察結束後小鼠全部處死取腦,冰凍切片行免疫組織化學染色後觀察Munc-18蛋白和c-Fos蛋白的錶達情況。結果註射海人藻痠30min後小鼠癲癇樣髮作,首先齣現鬍鬚和嘴部抽動,接著翹尾、甩尾、點頭、單側上肢抽搐,達到4級標準。與對照組和生理鹽水組比較,癲癇組小鼠海馬區域的Munc-18蛋白分佈減少同時伴有c-Fos蛋白的錶達增加,以CA3和齒狀迴區域更加明顯,差彆具有統計學差異。結論癲癇能夠導緻Munc-18蛋白在海馬區域分佈減少,同時使該區域的神經興奮性增高。

Objective Munc18-1 has an important role in neurotransmitter release which controls every step of release synap-tic vesicle.In the present study, whether epilepsy seizure causes a change of Munc-18 in hippocampus was investigated.Methods 18 KunMing mice were randomly divided into three groups:epilepsy group which was received intraperitoneal injection of Kainic acid (10mg/kg), saline group which was only received an injection of same amount of saline, control group which was received no treat-ment.Monitor the behavior of the mice, then the mice were slaughtered after investigation and the brain tissue was collected to perform frozen sections.Immunohistochemistry with anti-Munc-18 antibody was used to determine the distribution of Munc-18 protein in the hippocampus.Results 30 minutes after injection, behavioral changes of the mice appeared which including mouth and facial twit-ches, clonic head movements, unilateral forelimb clonus followed by contralateral clonus, clonic rearing.Compared with control group and saline group, the Munc-18 immunopositive cells were declined in CA3 and Dentate Gyrus of hippocampus;we also found that the expression of c-Fos proteins were significantly increased in the CA3 and Dentate Gyrus of hippocampus.Conclusion These results suggest that epilepsy seizures can induce the distribution change of Munc-18 in the hippocampus which may subsequently modulate neuronal functions in brain.
